摘要 目的:检测磷脂酰肌醇3-激酶(phosphoinositide 3-kinase,PI3K)/Akt信号转导通路中PI3K和磷酸化Akt在尖锐湿疣(CA)和生殖器鳞癌(SCC)组织中的表达。方法:应用免疫组化EnVision二步法对30例CA皮损、30例生殖器SCC和20例健康正常包皮组织中PI3K和p-Akt蛋白表达及分布进行检测。结果:PI3K蛋白在正常组织仅在基底层细胞表达;在CA皮损中颗粒层、棘细胞层和基底层细胞胞质呈不同程度阳性表达;在生殖器鳞癌细胞中呈不同程度阳性表达。p-Akt蛋白在正常组织仅在基底层细胞表达;在CA皮损中仅基底层散在树突状细胞呈阳性表达;在生殖器鳞癌肿瘤细胞中呈不同程度阳性表达。结论:PI3K/p-Akt该信号转导通路可能参与了鳞癌和CA的发生发展。 Objective: To investigate the roles of PI3K/p- Akt signal transduction pathway in pathogenesis of condyloma aeuminatum (CA) and the genital squamous cell carcinoma (SCC). Methods: The EnVision two steps method of immunohistochemistry was used to detect and compare the expression and distribution of PI3K and p - Akt protein in lesions of 30 cases of CA, 30 cases of genital SCC and 20 eases of normal foreskin. Results: The staining of PI3K was positive or strong positive in cytoplasm of the basal cell layer in normal foreskin. The expression of PI3K in cytoplasm of granular layer, prickle cell layer and basal cell layer in CA varied in extent. The expression of PI3K was positive or strong positive in tumor cell cytoplasm of genital SCC. The expression of p - Akt was positive or strong positive in the cytoplasm of the basal cell layer in normal foreskin. The prickle cell layer and granular layer of CA lesions did not express p - Akt, while the dendritic cells in the basal layer showed positive expression of p - Akt. The expression of p - Akt was weak in tumor cell cytoplasm and nucleus of the genital SCC tissues. Conclusion: PI3K/p- Akt cell signaling pathway may be involved in the development of SCC and CA.
