摘要
目的探讨MCF-7荷瘤小鼠模型血清和树突状细胞(dendritic cells,DCs)上清液中细胞因子水平的变化,为免疫治疗提供依据。方法裸鼠(对照组)及MCF-7裸鼠模型(荷瘤组)各10只,应用ELISA法检测血清及DCs上清液中肿瘤坏死因子(tumor necrosis factor,TNF)-α、白细胞介素(interleukin,IL)-6、IL-12水平。结果荷瘤组血清TNF-α水平((314.81±103.36)pg/mL)低于对照组((371.87±115.30)pg/mL),但2组比较差异无统计学意义(P>0.05),DCs上清液TNF-α水平((1.88±0.10)ng/mL)高于对照组((1.72±0.07)ng/mL)(P<0.05);荷瘤组血清IL-6水平((170.72±54.51)pg/mL)高于对照组((122.35±20.65)pg/mL)(P<0.05),DCs上清液IL-6水平((57.18±5.06)pg/mL)高于对照组((44.53±4.86)pg/mL)(P<0.05);荷瘤组血清及DCs上清液IL-12水平((119.80±33.53)pg/mL和(89.76±8.89)pg/mL)均低于对照组((186.22±51.91)pg/mL和(157.98±48.31)pg/mL)(P<0.05)。结论 MCF-7荷瘤小鼠血清IL-6可能参与该类乳腺癌肿瘤细胞的生长;DCs可能参与产生TNF-α,促进IL-6分泌,抑制IL-12分泌,对肿瘤细胞的杀伤作用减弱。
Objective To detect cytokines levels in serum and dendritic cells (DCs)-derived supernatans in MCF 7 mice in order to provide a basis for immunotherapy. Methods The levels of tumor necrosis faetor-a (TNF a), interleukin (IL)-6 and IL-12 were detected serum and DCs derived supernatans with ELISA in 10 MCF-7 mice (tumor-bearing mice) and 10 nude mice. Results The level of TNF-a in serum was (314. 81± 103. 36) pg/mL in tumor bearing mice and (371.87±115.30) pg/mL in nude mice, which showed no significant difference (P〉0.05). And the level of TNF-a in DCs-derived supernatans was (1.88 ± 0. 10)ng/mL in tumor-bearing mice and (1. 72 ± 0. 07) ng/mL in control mice, which showed a significant difference (P^0.05). The IL-6 levels in serum and DCs-derived supernatans were (170.72± 54. 51) pg/mL and (57. 18 ± 5. 06) pg/mL in tumor-bearing mice, significantly higher than those in nude mice ((122.35±20. 65) pg/mL and (44. 53±4. 86) pg/mL) (P〈0.05). The IL-12 levels in serum and DCs-derived supernatans were (119.80± 33.53) pg/mL and (89.76 ± 8.89) pg/mL in tumor bearing mice, significantly lower than those in nude mice ((186.22-1-51.91) pg/mL and (157.98-48.31) pg/mL) (P〈0.05). Conclusion IL 6 in serum may participate breast cancer cell growth in MCF-7 tumor-hearing mice. DCs may participate producing TNF-a, promoting the secretion of IL-6 and inhibiting the secretion of IL 12, and may decrease the ability of killing tumor cell.
