摘要
目的探讨树突状细胞(dendritic cell,DC)在小鼠移植瘤中的分布和形态学特征。方法采用光镜、透射电镜和免疫组织化学方法观察30只小鼠移植瘤中树突状细胞分布及形态。结果树突状细胞主要分布在小鼠移植瘤周边区及瘤巢内,实验组树突状细胞的数量比对照组明显增多(P<0.01)。电镜下,树突状细胞体积大,形态不规则,表面有许多树枝状突起,核不规则,有切迹。胞浆内有丰富的线粒体、核糖体和内质网。树突状细胞具有两种形态,一种表现为突起多,细胞器丰富;另一种表现为突起较少,细胞器也较少。树突状细胞与瘤细胞、树突状细胞与淋巴细胞、淋巴细胞与瘤细胞之间存在多种接触形式。同时可见淋巴细胞与淋巴细胞,树突状细胞与树突状细胞之间也有紧密接触。还可见树突状细胞与瘤细胞接触,接触部位有颗粒物质,瘤细胞趋于凋亡状态。结论树突状细胞在小鼠移植瘤组织中有两种类型,且在肿瘤组织中数量显著高于正常组织。
Objective To explore the morphological characteristics and distribution of dendritic cells (DC) in transplanted tumor of mice. Methods The morphology and distribution of DC in transplanted tumor of 30 mice were observed by light microscopy, electron microscopy and immunohistochemistry. Results DCs were found mainly in the tumor nests and tissue surrounding tumor. There were more DCs in experimental group than those in control group (P 〈 0.01 ). Under electron microscope, DCs were bigger and irregular in shape with many dendritic protrusions, irregular nucleus, large amount of mitochondria, ribosomes, Golgi complexes and endoplasmic reticulums. Two types of DCs were found. One of them contained more dendritic protrusions and abundant organelles. The other contained less dendritic protrusions and lacked organelles. Various contacts were seen between DC and tumor cells, DC and lymphocytes, lymphocytes and tumor cells. The intimate contacts were also found between lymphocytesand lymphocytes, DC and DC. In addition, contacts were also seen between DCs and tumor cells which showed apoptotic features. Conclusion Two types of DCs 'exist in transplanted tumor of mice,there are more DCs in tumor tissues than in normal tissues.
