蒲公英总黄酮对假单胞菌抑菌机理的探讨 被引量:16

摘要 对蒲公英总黄酮进行提取和抑菌实验。以革兰氏阴性假单胞菌做为试验菌,通过测定蒲公英总黄酮类提取物与假单胞菌的作用前后细菌培养液的电导率、可溶性总糖及蛋白质表达方面的变化,初步弄清了蒲公英总黄酮类提取物对假单胞菌的抑菌机理。研究结果表明,经过蒲公英总黄酮类提取物处理后,细菌培养液的电导率和可溶性总糖的浓度都增加,表明蒲公英总黄酮类提取物能破坏细胞膜的结构、导致细胞通透性增加,使细胞内容物外泄。另外,通过实验证实蒲公英总黄酮类提取物可以防碍细菌蛋白质的正常表达,导致细菌正常生理功能丧失,起到抑菌作用。 This is a experiment about investigating the bacteriostatic effects of extracts of total flavonoids from Taraxacum mongolicum Hand-Mazz. The antimicrobial mechanisms of extractions of total flavonoids from Taraxacum mongolicum Hand-Mazz against Gram-negative pseudomonad were investigated by determining the changes in electric conductivity, total sugar concentration and protein expression of bacteria. The facts that the electric conductivity and total sugar concentration of microbial broth increased indicated that extractions of total flavonoids from Taraxacum mongolicum Hand-Mazz could damage the structure of cell membrane, which resulted in the increase of permeability of cell membrane and release of cell components. In addition, the experiments showed that the extractions could block the protein expression in bacteria and led to lost of normal physiological function of bacterium, which finally exerted a role of antimicrobial.
