摘要
确定葛花中总黄酮提取的优化工艺条件,并对提取物进行含量测定和体外抗氧化作用研究。通过正交试验,确定葛花中总黄酮的最佳工艺条件为:50%甲醇溶液,料液比为1∶30(g/mL),在70℃温度下,超声提取2.0 h,葛花中总黄酮的含量为17.48%。以ABTS.+、DPPH、NO等检测其清除自由基活性。结果表明,葛花总黄酮具有一定的清除DPPH、NO、O2-.能力和一定的抗氧化能力。
To identify the optimal extraction process of total flavonoids from Puerariae Flos, determine its content and investigate its free radical scavenging effect in vitro. With orthogonal test, the optimal conditions were determined as follows : methanol concentration of 50 %, temperature of 70 ℃, extraction time of 2.0 h, and ratio of solid to liquid was 1:30 (g/mL). Under these conditions, the extraction yield was 17.48%. The scavenging free radical activities, including ABTS·+, DPPH, NO and O2-· radicals were studied. The results indicated that the total flavonoids from Puerariae Flos exhibited free radical scavenging activity against DPPH, NO, O2-· and antioxidant capacity to some extent.
