摘要
研究益母草中多糖的最佳提取工艺并测定含量。以益母草为原料,蒸馏水为提取液,用超声波提取益母草中的多糖,并利用苯酚-硫酸法测定益母草中多糖的含量。进行单因素试验,考察提取时间、料液比、提取温度对益母草中多糖得率的影响,并通过正交试验设计来优化益母草中多糖的提取工艺,以确定其最佳的提取工艺。其最佳提取工艺条件为:提取时间为15 min,料液比为1∶15,提取温度为35℃,在最佳工艺条件下多糖的提取率为3.24%,平均回收率为93.04%(n=5)。
The experiment study on optimizing the extraction process for the Leonurus Japonicus Houtt of polysaccharide and determining the contents . The experiment ehoosed the Leonurus Japonicus Houtt as raw material and the distilled water as extracting solution. Using the method of ultrasonic extracted the polysaccharide. The contents determined by the phe-nol-vitriolic eolorimetry,by means of studying on the single factors extraction time, the solid to liquid , extraction temperature influence on the extraction ratio, and optimizing the extraction process for the Leonurus Japonicus Houtt of polysaccharide by orthogonal experimental design, confirmed that the optimum extraction . the optimum extraction as follows: extraction time was 15 minutes, the solid to liquid was 1:15, extraction temperature was 35 ℃. On the bases of the optimum extraction,the extraction rate for the Leonurus Japonicus Houtt of polysaccharide was 3.24 %. The average of recoveries was 93.04 % (n=5).
