摘要
采用AFLP分子标记技术对57份白榆无性系进行遗传多样性和亲缘关系的分析。8对引物对57份白榆无性系扩增后检测出1092条谱带,其中有1065条多态带,多态带比例为97.53%,表明所试白榆材料具有丰富的遗传多样性。遗传相似性系数为0.4266~0.7989,平均值为0.6114,说明各无性系之间有一定的遗传分化。聚类分析将白榆种质分成11组,来源相同的无性系并未严格聚在一起,表明白榆无性系的变异与地理来源没有严格的相关性。该项研究为更为深入地了解白榆遗传背景和遗传结构提供了参考,也为白榆种质资源的利用提供了依据。
The genetic diversity of 57 Ulmus pirnila L. clones are tested by AFLP. 8 primer combinations selected from 64 primer combinations produce a total of 1092 fragments, 1065 of which are polymorphic with a polymorphism percentage of 97.53%. It suggests that Ulmus pimila L. has a rich genetic diversity. The molecular genetic similarity coefficients rang from 0. 4266 to 0. 7989, and that the average being 0. 6114indicates that genetic variation occurs among them. All of Ulmus pimila L. are clustered into 11 groups with UPGMA, and the clones originated the same place don't distribute in the same group total- ly. It indicates that there is no strict relationship between the variation and geographic origin of the black pine clones. This study provides the reference for the genetic background and the genetic structure of Ulmus pirnila L. , and also provide the ba- sis for the utilization of the germplase of Ulmus pimila Linn..
