摘要 目的研究芒果苷对肺癌A549细胞增殖与凋亡的影响,为深入研究芒果苷抗肿瘤机制提供试验基础。方法 0、50、100、150、200、250μmol.L-1芒果苷处理肺癌A549细胞24或48 h后,采用四甲基偶氮噻蓝法检测芒果苷对A549细胞增殖的影响;采用流式细胞仪AnnexinV-异硫氰酸荧光素/碘化丙啶双染法检测芒果苷作用后细胞凋亡情况;荧光显微镜观察芒果苷作用后细胞形态变化。结果随着芒果苷浓度的增加和作用时间的延长,对A549细胞的增殖抑制作用增加,呈现明显的时间-剂量效应关系;流式细胞检测细胞凋亡分析表明,随着芒果苷浓度的增高,A549细胞早期凋亡率和晚期凋亡率均逐渐增加,与0μmol.L-1组比较差异有统计学意义(P<0.05);荧光电子显微镜观察芒果苷作用后的细胞中可见核固缩、核碎裂等典型的凋亡改变。结论芒果苷在体外能有效抑制肺癌细胞生长,其机制与其诱导肺癌细胞凋亡有关。 Objective To study the effects of mangiferin on the proliferation and apoptosis of lung cancer A549 cells in order to supply experimental basis for the further invesitigation of anti-tumor mechanism of mangiferin.Methods A549 cells were treated with 0,50,100,150,200 and 250 μmol·L-1 mangiferin for 24 or 48 hours.The effect of mangiferin on the proliferation and apoptosis of A549 cells was determined by methyl thiazolyl tetrazolium assay and flow cytometric analysis with AnnexinV-fluorescein isothiocyanate/propidine iodine respectively;the changes of cell morphology was observed by fluorescent microscopy.Results With the increasing of the concentration of mangiferin and the extension of time,the inhibition of mangiferin on proliferation of A549 cells increased,it showed that the mangiferin inhibited cell proliferation in A549 cell lines in a dose-and time-dependent manner;the results of flow cytometric analysis showed that early and late apoptosis rates were gradually increased with the concentration of mangiferin increasing,there were significant differences compared to 0 μmol·L-1 group(P0.05).As demonstrated by the fluorescent microscopy results,treatment with mangiferin significantly induced the apoptotic cell death with chromatin condensation and nuclear fragmentation.Conclusion Mangiferin can inhibit the proliferation of A549 cell lines in vitro through inducing the apoptosis of A549 lung cancer cell.
