摘要
目的探讨薯蓣皂素(Diosgenin)对人视网膜色素上皮细胞系(ARPE-19)的增生、迁移的影响。方法用不同浓度(5,10,20,30,40,50,60,70μg/ml)的薯蓣皂素处理ARPE-19细胞24 h,空白组(0μg/ml)作对照,MMT检测细胞增生情况并确定药物的安全浓度范围,观察安全浓度薯蓣皂素48 h和24 h对RPE细胞的增殖抑制作用。以安全浓度的薯蓣皂素利用Transwell小室检细胞检测迁移能力变化。结果薯蓣皂素可抑制ARPE-19细胞的增生、迁移,并随药物浓度增加抑制作用增强,药物浓度在20μg/ml范围内是安全有效的,与对照组相比有统计学意义(P<0.05),安全浓度药物作用24、48 h结果无显著性差异。结论薯蓣皂素能有效抑制ARPE-19细胞的增殖、迁移,可作为一种新的候选药物抑制增生性玻璃体视网膜疾病(proliferative vitreoretinopathy,PVR)的发生发展。
Objective To investigate the effects of Diosgenin on the proliferation and migration of human retinal pigment epithelial cell (RPE) line (ARPE- 19). Methods ARPE- 19 cells were treated by different concentration (5,10,20,30,40,50,60,70 μg/ml) of diosgenin 24 hours, while the control group processed without diosgenin(0 μg/ml). MMT assay was utilized to test cell proliferation, confirm the safe concentration of the drug, and observe the the inhibitive effects of safe concentration diosgenin on RPE cells at 24 h and 48 h. Different safe concentra- tion drugs were adopted to investigate cell migration through Transwell unit. Results Diosgenin inhibited proliferation of RPE cells in a dose-dependent manner. When treated with non-toxic doses of diosgenin, cell migration were markedly suppressed by in vitro Transwell unit assay, respectively. Inhibition increased with the increase of drug concentration, the safe and effec- tive drug concentration within the 20 μg/ml, compared with the control group was statistically significant (P 〈0.05). Conclusion Diosgenin can effectively inhibit the ARPE-19 cell proliferation and migration, these findings reveal that diosgenin might be a new therapeutic potential drug for preventing proliferative vitreoretinopathy (PVR).
