摘要
目的 探讨白细胞介素 1(IL 1)对骨髓瘤细胞粘附诱导骨髓基质细胞 (BMSCs)分泌白细胞介素 6(IL 6 )的影响及多发性骨髓瘤 (MM )患者和正常对照BMSCs分泌IL 6的差异。方法 建立人BMSCs培养方法 ,采用MTT比色法检测BMSCs培养上清中IL 6量。结果 (1)MM患者BMSCs具有异常增殖能力 ;(2 )单纯KM 3细胞培养上清中IL 6量为 7± 2 .3u/ml;(3)单纯BMSCs培养时 ,MM患者BMSCs分泌IL 6量为 2 6±7.5u/ml,高于正常对照 16± 4.7u/ml(P <0 .0 1) ;(4)KM3细胞与MM患者或正常对照BMSCs共培养 2 4小时后 ,其培养上清中IL 6量分别为 49± 18.1u/ml和 2 7± 6 .9u/ml,均高于单纯BMSCs培养上清中IL 6量 (P<0 .0 1) ;(5 )将BMSCs经 1%多聚甲醛固定 ,再与KM 3细胞共培养 2 4小时后 ,其培养上清中IL 6量与单纯KM3细胞培养上清比较差异无显著性 ;(6 )单纯BMSCs培养体系中加入IL 1,IL 6分泌量差异无显著性 ;(7)在KM 3细胞与BMSCs共培养体系中加入IL 1,IL 6分泌量达 74± 2 0 .5u/ml(MM组 )和 46± 10 .5u/ml(正常对照组 ) ,均高于单纯KM 3与BMSCs共培养 (P <0 .0 1)。结论 MM患者BMSCs具有异常增殖和分泌IL 6能力 ,IL 1通过骨髓瘤细胞介导可促进BMSCs分泌IL
Objective To clarify the effect of adhesion on IL 6 secretion by BMSCs and the difference of IL 6 secretion by BMSCs between MM and normal controls. Methods Establishing culture method of human bone marrow stromal cells (BMSCs), IL 6secretion by BMSCs was detected by MTT colorimetry. Results IL 6 activity detected in the supernatants of KM3 was 7±2.3 u/ml, KM3 could secrete IL 6 to maintain its proliferation. Under normal condition, the amount of IL 6 secreted by BMSCs of MM was higher than that of normal control (26±7.5 u/ml vs 16± 4.7 u/ml, P <0.01). KM3/BMSCs of MM or normal control co cultured, after 24 hours, the amount of IL 6 of supernatant (49±18.1 u/ml or 27±6.9 u/ml) was much higher than that of pure BMSCs culture (26±7.5 u/ml or 16±4.7 u/ml P <0.01). BMSCs was fixed with 1% paraformaldehyde before adherence of KM3 cells, compared to he supernatants of KM3, no significant increase in IL 6 was noted. When IL 1 was added to pure BMSCs culture, secretion of IL 6 did not change significantly, but when IL 1 was added to KM3/BMSCs of MM or normal control co culture system, secretion of IL 6 increased significantly (MM 74±20.5 U/ML; normal conrol 46±10.5 u/ml). Conclusion BMSCs of MM have abnormal proliferative capacity and secretory characteristics. IL 1 can increase myeloma cells adhesion induced IL 6 secretion by BMSCs. (Shanghai Med J, 2000,23∶362 364)
