摘要
目的探讨肝硬化患者血清IL-8含量及外周血单个核细胞(PBMCs)中IL-8 mRNA表达,及其与ALT、AST相关性。方法以ELISA法检测血清IL-8含量,定量PCR检测65例肝硬化患者(代偿期36例,失代偿期29例)外周血IL-8 mRNA水平,设GAPDH为参照,以logcDNA/logGAPDH比值代表其最终mRNA水平。结果肝硬化患者外周血IL-8及其mRNA表达量分别为(448.41±59.46)pg/mL、(1.4850±0.1801)logcDNA/logGAPDH,较正常组显著增高,差异有统计学意义(P<0.01)。其中,ALT异常组IL-8及其mRNA表达量分别为(465.73±48.88)pg/mL、(1.5207±0.1684)logcDNA/logGAPDH,与ALT正常组相比,差异有统计学意义(P<0.01)。AST异常组IL-8及其mRNA表达量分别为(467.39±48.48)pg/mL、(1.5419±0.1650)logcDNA/logGAPDH,与AST正常组相比,差异有统计学意义(P<0.01)。肝硬化外周血IL-8及其mRNA与ALT(r=0.764,r=0.612,P<0.01)及AST(r=0.711,r=0.579,P<0.01)显著相关。结论肝硬化外周血IL-8及其mRNA表达升高,并与血清ALT、AST水平密切相关,提示IL-8可能参与了肝硬化的致炎分子病理过程,似可作为反映肝脏炎症变化的新型辅助指标。
Objective To study the expressions of IL-8 in serum and its mRNA in peripheral blood as well as their correlation with ALT and AST in the sera of cirrhosis patients.Methods The levels of IL-8 in the sera of sixty-five cirrhosis patients(compensated cirrhosis 36,decompensated cirrhosis 29) were detected by ELISA,and the mRNA expression of IL-8 mRNA in PBMCs was examined by Real-time PCR.The ratio of logcDNA/logGAPDH was regarded as the extreme level of IL-8 mRNA.Results The levels of IL-8 and its mRNA in peripheral blood of the patients were(448.41±59.46) pg/mL and(1.4850±0.1801) logcDNA/logGAPDH,which were significantly higher than those in controls(P0.01).The levels of IL-8 and its mRNA in ALT abnormal group were(465.73±48.88) pg/mL and(1.5207±0.1684) logcDNA/logGAPDH,which were significantly higher than those in ALT normal group(P0.01).The expressions of IL-8 and its mRNA in AST abnormal group were(467.39±48.48) pg/mL and(1.5419±0.1650) logcDNA/logGAPDH,and were also higher than those in AST normal group(P0.01).The expressions of IL-8 and its mRNA were obviously correlated with the levels of ALT(r=0.764,r=0.612,P0.01) and AST(r=0.711,r=0.579,P0.01) in cirrhosis.Conclusion The high levels of IL-8 and its mRNA expression in cirrhosis are closely associated with the degrees of ALT and AST in serum,which suggests that IL-8 may have played a key role in the pathological process of cirrhosis,and may be regarded as a new ancillary indicator for hepatocytes injury,and can play a key role
