摘要
在用体内-体外结合的方法建起了一株食管癌前恶性转化细胞系RE_(26-3)的基础上,用同样的方法建立了另一株不具备恶性转化的癌前细胞系RE_(46).提示两者病理形态上均为不典型增生,通过体外培养RE(46)细胞生物学上只有癌前细胞的特性不具恶性转化潜能,不能继续传代,而RE_(26-3)逐步显示各种恶性转化细胞的生物学特征,建成癌细胞系.
With the use of the same method, a neoplastic transformation cell line (RE25-3) and a non-malignant cell line (RE46) of esophageal epithelium of rat have been established in our laboratory. Comparison studies of the difference of their biological characteristics have been performed. The experimental results showed that the difference of biological characteristics of these two cell lines was significant. The mitotic index of RE25-3 was 2. 0% while that of RE46 was 1. 38%. The number of chromosome in RE25-3 was higher than that of RE46, the tetraploid in RE25-3 was 56. 44% while in RE46 was just 5. 55%. The micronuclei rate of the RE25-3 was also higher than that of the RE46. The RE25-3 cells could be cultured in soft agar while the RE46 could not. RE25-3 could be subcultured for more than 50 generations while RE46 only could be subcultured for seven generations. While using the morphological criteria does not answer the question whether neoplastic trans-formation potency exists in these two ceil lines , our observation shows that definite neoplastic characteristic changes of genetic substances do exist prior to the appearance of morphological changes.
