麦冬多糖对2型糖尿病大鼠肾脏的保护作用 被引量:19

摘要 目的观察麦冬多糖对2型糖尿病大鼠肾脏的保护作用。方法采用链脲佐菌素(STZ)诱导2型糖尿病大鼠模型,给予不同剂量麦冬多糖(200、400、800 mg/kg)治疗6周后,检测各组大鼠空腹血糖(FBG)、肾脏指数、血肌酐(Scr)、尿素氮(BUN)含量,比较各组大鼠肾组织中总抗氧化能力(T-AOC)及过氧化氢(H2O2)的含量,Western bolt检测肾组织中p47phox、核因子-κB(NF-κB)蛋白表达。结果模型组大鼠FBG、肾脏指数、Scr、BUN含量明显升高,肾脏T-AOC明显降低,肾组织p47phox、NF-κB的表达及H2O2含量显著升高。麦冬多糖能明显降低糖尿病大鼠FBG、肾脏指数、Scr及BUN水平,提高肾脏T-AOC,降低肾脏p47phox、NF-κB的表达及H2O2的含量。结论麦冬多糖对2型糖尿病大鼠肾脏具有一定的保护作用,其机制可能与其抗氧化、下调糖尿病大鼠肾脏p47phox、NF-κB的表达有关。 Objective To observe effect of ophiopogon-polysaccharide on protection of kidney in type 2 diabetic rats. Methods Streptozotocin-induced diabetic rats were given ophiopogon-polysac- charide (200,400, 800 mg/kg) for 6 weeks. The changes in blood glucose (FBG), kidney weight index, and serum creatinine (Scr) and blood urea nitrogen (BUN) were determined after treatment. The total antioxidant capacity (T-AOC) and hydrogen peroxide (H202 ) in renal tissue were measured and compared between groups. In addition, the expressions of p47^phox, nuclear factor-KB (NF-KB) in the kidney tissues were determined by western bolt. Results BG, kidney weight index and Scr and BUN in the model group increased significantly, renal T-AOC significantly decreased. The contents of renal H202 and expressions of renal p47^phox, NF-KB increased significantly. Compared with model group, the level of BG, kidney weight index and Scr and BUN in ophiopogon-polysaccharide groups decreased significantly, and the renal T-AOC increased significantly and the content of renal H202 and the expression of renal p47^phox, NF-KB decreased significantly. Conclusion Ophiopogon-polysaccha- ride can alleviate the renal lesions in type 2 diabetic rats. Antioxidation and inhibition of renal p47^phox, NF-KB expressions maybe the possible mechanism for protection of kidney.
