丙泊酚与东莨菪碱合用对大鼠认知功能的影响 被引量:1

摘要 目的观察丙泊酚与东莨菪碱合用对大鼠认知功能的影响。方法健康成年雄性SD大鼠56只,体重200-250 g,随机分为4组,每组14只:实验药品均采用腹腔注射,对照组(等体积生理盐水)、丙泊酚组(100 mg/kg)、东莨菪碱组(0.8mg/kg)、丙泊酚+东莨菪碱组(100 mg/kg+0.8 mg/kg)。观察翻正反射。随机选取每组8只大鼠在注射丙泊酚12 h后进行Morris水迷宫测试,另外6只进行跳台测试。用水迷宫实验和跳台实验分别观察大鼠逃避潜伏期和错误次数。结果丙泊酚组和丙泊酚+东莨菪碱组的翻正反射消失时间及翻正反射恢复时间差异无统计学意义。水迷宫测试表明,与对照组相比,东莨菪碱组第1-4天潜伏期均延长(P<0.05-0.01),且第5天目标象限时间百分比明显减少(P<0.05),而丙泊酚组则在第1-2天潜伏期延长(P<0.01),第3-4天的潜伏期和第5天时间百分比差异无统计学意义(P>0.05);与东莨菪碱组比较,丙泊酚+东莨菪碱组第1-2天潜伏期延长(P<0.05),第3-4天的潜伏期和第5天时间百分比差异无统计学意义(P>0.05)。跳台实验中,与对照组相比,东莨菪碱组和丙泊酚组第1-2天的潜伏期缩短(P<0.01),错误次数增多(P<0.05-0.01),第3-4天东莨菪碱组的潜伏期缩短(P<0.01),错误次数增多(P<0.05),而丙泊酚组差异无统计学意义;与东莨菪碱组相比,丙泊酚+东莨菪碱组第1天潜伏期缩短(P<0.01),错误次数增多(P<0.01),第2天潜伏期略缩短(P>0.05),错误次数增多(P<0.05),第3-4天不论是错误次数,还是潜伏期均相似(P>0.05)。结论单独使用丙泊酚和东莨菪碱均可导致大鼠学习记忆障碍;两药合用对大鼠认知功能的影响只呈短暂协同作用,而远期无影响。 Objective To explore the effect of propofol combined with scopolamine on cognitive function in rats.Methods Fifty-six SD rats weighing 230-250 g were randomly divided into 4 groups:control group(saline,n=14),propofol group(100 mg/kg,n=14),scopolamine group(0.8 mg/kg,n=14) and propofol+scopolamine group(100 mg/kg+0.8 mg/kg,n=14).The duration of loss and recovery of righting reflex was observed.Eight rats in each group underwent Morris water maze(MWM) test for 5 consecutive days and the other 6 rats performed step-down test from 12 h after the injection of propofol.MWM test and step-down test were conducted to evaluate the effects of propofol on learning and memory function in terms of escape latency and numbers of error.Results There was no significant difference in the duration of loss and recovery of righting reflex between propofol group and propofol+scopolamine group.In MWM test,compared with control group,the spatial learning at day 1-4 and memory of rats at day 5 was impaired in scopolamine group(P0.05-0.01).The mean escape latency was significantly larger at day 1-2(P0.01) and no difference in the mean escape latency at day 3-4 and the swimming time percentage in platform quadrant at day 5 was found in propofol group(P0.05).Compared with scopolamine group,memory disorders increased at day 1-2 in propofol+scopolamine group(P0.05),and there was no significant difference in learning and memory function test at day 3-5(P0.05).In step-down test,as compared to that in control group,the latencies were shortened at day 1-2(P0.01) and the numbers of error were increased(P0.05-0.01) in propofol group and scopolamine group,and they had the same trend at day 3-4 in scopolamine group(P0.05) but not in propofol group.Compared to scopolamine group,the latency was decreased at day 1(P0.01) and the numbers of error were increased at day 1-2 in propofol+scopolamine group(P0.05),however,there was no significant difference at day 3-4(P0.05).Conclusion Propofol and scopolamine used alone can both lead to the rat memory disorders.Propofol can aggravate transiently the cognitive dysfunction induced by scopolamine in rats.
