摘要
目的分析趋化性细胞因子CCL3L1基因拷贝数在中国汉族健康人和HIV-1感染者中的分布特点,以期阐明趋化性细胞因子CCL3L1基因拷贝数与HIV-1易感性的相关性。方法选择HIV/AIDS患者81例和健康者97例,实时定量PCR法检测趋化性细胞因子CCL3L1基因拷贝数。bDNA法检测HIV/AIDS患者的血浆病毒载量,同时用流式细胞仪检测外周血CD4+T细胞数量。结果中国汉族健康人群的CCL3L1平均基因拷贝数为1,应用logistic回归分析发现低于汉族健康人群平均拷贝数者有较高的易感性(OR值为0.501);应用spearman相关分析,HIV/AIDS人群CCL3L1基因拷贝数与病毒载量呈密切负相关性(P=0.000),HIV/AIDS人群CCL3L1基因拷贝数与CD4T细胞计数密切正相关(P=0.000)。结论趋化性细胞因子CCL3L1基因拷贝数与HIV-1易感性密切相关。
Objective To detect the human chemokine CCL3-L1 gene copy numbers of healthy and HIV-1infected in dividuals of Han Chinese population,and to evaluate the correlation between CCL3L1 gene copy numbers and the risk of acquiring HIV-1 infection.Methods Case control study was adopted,gene copy numbers of the human chemokine CCL3L1 from 178 samples were detected by real time polymerase chain reaction.81 samples were detected the initial viral load by HIV-1 Quantiplex(bDNA) and CD4+ T cells by flow cytometry in the HIV positive cohort.Results CCL3L1 median gene copy number of Chinese han population was one.We used logistic regression to estimate the risk of acquiring HIV-1 associated with category of CCL3L1 gene copy number by comparing it with a reference category which was the population-specific median gene copy number.Possession of a CCL3L1 copy number lower than the population average is associated with markedly enhanced HIV-1 susceptibility(OR=0.501).The spearman test showed that there was significant negative correlation between different CCL3L1 gene doses and the initial viral load in the HIV-positive cohort(P=0.000).The spearman test showed that there was significant positive correlation between different CCL3L1 gene doses and CD4+ T cells count in the HIV-positive cohort(P=0.000).Conclusion The gene copy numbers of the human chemokine CCL3L1 might be associated with susceptibility to HIV-1 infection.Further studyof the implicated in needed.
