摘要
目的 研究TIMP 2基因转染胃癌细胞后的生物学行为变化。方法 采用基因转染的方法将人全长TIMP 2cDNA基因转染入人胃癌SGC 790 1细胞系中 ,利用层粘连蛋白 (LN)对肿瘤细胞的诱导迁移作用 ,在体外侵袭模型中观察转染基因细胞侵袭能力的改变。结果 转染TIMP 2基因的SGC 790 1细胞侵袭能力 [33 1± 11 3细胞 h]明显低于未转染组 [( 135 7± 2 1 6 )细胞 h ,P <0 0 1],肿瘤细胞在LN上的粘附能力受到抑制 ,转染组 ( 417 7± 2 1 5 )低于未转染组 ( 82 9 4± 45 7) (P <0 0 5 )。且细胞的增殖能力受到限制。结论 TIMP 2基因可阻抑层粘连蛋白对瘤细胞的诱导侵袭影响 。
Objective To study the change of human gastric cancer cells SGC7901 transfected by tissue inhibitor metalloproteinase2 (TIMP2) gene in experimental laminin reaction. Methods TIMP2 gene was transfected into human gastric cancer cells SGC7901 by Liprofectimin technique. A model of tumor invasion in vitro was used to determine the malignant behavior of tumor cells. The transfected and untransfected groups were observed for adhesion、invasion、migration and proliferation of SGC7901 cells on laminin. Results The invasion ability of SGC7901T2 (33.1±11.3 cells/h) was significantly lower than the controls (135.7±21.6 cells/h,P<001). TIMP2 could inhibit adhesion、invasion、migration and proliferation of SGC7901 cells on laminin.Conclusion The inhibitory effects of TIMP2 on cancer metastasis related with laminin. It shows a key action in blocking cancer metastasis.$$$$
