止血带对克林霉素在兔骨组织中分布的影响

摘要 目的:建立高效液相色谱法测定兔骨组织中的克林霉素,并对其在使用止血带后,在骨组织的分布特征进行研究。方法:以乙腈-四氢呋喃-磷酸盐缓冲溶液(30∶1∶70,pH 5)为流动相,柱温40℃;流速0.8mL·min-1;以替硝唑为内标,以LunaC18柱(250mm×4.6mm,5μm)进行分离,检测波长为205nm。测定静脉注射克林霉素(20mg·kg-1)后30,60,90,120min(A、B、C、D)上止血带分别阻断血液循环30,60,90min实验组与未止血对照组的骨组织中药物含量。结果:克林霉素在10.58~264.5μg·mL-1范围内线性关系良好,日内RSD≤2.17%,日间RSD≤2.99%。实验组克林霉素含量与对照组比较,差异有极显著性(P<0.01);相同止血时机,不同止血时间30,60,90min组之间比较,无显著性差异(P>0.05);不同止血时机组,A组与B、C、D组比较差异有极显著性(P<0.01),B、C、D组骨组织的克林霉素含量较高,3组之间差异无显著性(P>0.05)。结论:方法准确可靠、简便、重现性好,上止血带组骨组织中克林霉素含量明显低于无止血带组,止血时机是影响克林霉素骨组织分布的主要因素。 OBJECTIVE To establish a HPLC method for determining the content of the clindamycin in bone tissue of rab- bits, and study its characteristics of distribution in bone tissue of rabbits which were stopped bleeding by tourniquet. METHODS The mobile phase consisted of a mixture of acetonitrile tetrahydrofuran- phosphoric acid(30: 1:70, pH = 5) with a flow rate of 0. 8 mL-min-1 ; Tinidazole was used as an internal standard, the analytical column was packed with 5μm C18 at 40℃ ;and the detection wavelength was 205 nm. Experiment groups were stopped bleeding for 30,60,90 min after administration of clinda- mycin 30,60,90,120 min(A, B, C, D) respectively, and then the contents of the clindamycin in bone tissue of rabbits of experi- ment and control groups were determined by HPLC. RESULTS The calibration curve of clindamycin was good linear in the range of 10. 58~264. 5μg ·mL^- 1. The RSD of intra-day and inter-day was no more than 2. 17% and 2. 99% respectively. The concentrations of clindamycin in experiment groups compared with those in control groups were significant differences (P〈 0. 01 ) ;in the same stopped bleeding occasion groups, compared with different stopped bleeding time iength groups there was no significant difference (P〈0. 05);in different stopped bleeding occasion groups, there was significant difference between A and B, C, D respectively; the contents of clindamycin in groups of B, C, D were higher, and there was no significant difference among B,C,D groups. CONCLUSION This method is accurate, reliable, simple and reproducible. The content of clindamycin under tourniquets is lower than that without tourniquet significantty in bone tissue. Stop bleeding occasion is the principal factor to affect the distribution of clindamycin in bone tissue.
