摘要
化疗药物是临床治疗肿瘤的常见方法,但其先天性和获得性耐药也是临床亟需解决的问题。肿瘤细胞DNA修复能力的改变是引起该类药物产生耐药的重要因素之一。DNA修复是维持基因组稳定的主要因素,一种DNA损伤可能被多种DNA修复通路修复。了解DNA修复与化疗药物耐药性之间的关系,开发DNA修复抑制剂联合化疗可以克服化疗药物的耐药性并提高临床效果,同时为临床合理运用化疗药物及研制低耐药性药物提供理论依据。
chemotherapy is a common treatment for cancer,but its natural and acquired resistance is a serious problem that limits its clinical application.The change of DNA repair ability of cancer cells is one of the reasons which cause resistance to chemotherapeutic agents.DNA repair system is the main factor to maintain the genome stability,usually several repair pathways are involved in the chemotherapeutics efficacy.Therefore,it is important to understand the relationship between DNA repair and chemotherapeutic drug resistance,which can provide the theoretical basis for the clinical use of chemotherapeutic agents and developing new anticancer agents which can overcome the drug resistance.
