柯萨奇B3病毒引起心肌细胞凋亡的初步探索 被引量:6

Preliminary study on myocardial cell apoptosis induced by Coxsackie virus B3

摘要 目的 以病毒性心肌炎 (viralmyocarditis,VMC)细胞感染模型为基础 ,研究柯萨奇B3病毒 (CoxsackieB3virus,CVB3)感染与心肌细胞凋亡的关系。方法 在嗜鼠心肌柯萨奇B3病毒(CVB3m)感染心肌细胞后 2 4h电镜观察细胞超微结构的改变 ,同时取细胞悬液以流式细胞仪检测“凋亡峰” ,了解细胞凋亡率 ,并采用酶联免疫分析法 (ELISA)定量测定CVB3m病毒感染后 1、2、4、7、2 4h心肌细胞胞浆内组蛋白伴随DNA片段 (单核小体和寡核小体 )的多少 ,进一步了解凋亡程度。结果 电镜观察CVB3m感染的心肌细胞的超微结构符合凋亡改变 ,流式细胞仪DNA倍体分析检出凋亡峰 ,ELISA法检测提示病毒感染后出现心肌细胞凋亡 ,且在一定时段内凋亡程度随时间延长而增高。结论CVB3m病毒感染可诱发心肌细胞凋亡 ,且随时间延长 ,凋亡量增多 ,凋亡程度加重 ,凋亡可能参与VMC的病理过程。 Objective To determine the relationship between CVB3's challenge and apoptosis in myocardial cells. Methods The micro model of cultured new born rat heart cells infected by CVB3m was established. To observe the apoptotic cell's morphologic change by EM, measure “apoptotic peak” by flow cytometry, and detect histone associated DNA fragmes by ELISA. Results Morphologic features showed that aggregation of chromatins into denser masses lying at the periphery of the nucleus was observed by EM, “apoptotic peak” was detected by flow cytometry, and the photometric enzyme immunoassay revealed strong positive signals of apoptosis in CVB3m infected cells. Conclusion The CVB3m' infection can induce apoptosis in rat cardiomyocytes.
