摘要
研究了CHO细胞连续传代培养对细胞毒性试验结果的影响。对不同代次CHO细胞形态进行观察,并利用中性红比色法、MTT比色法检测同种受试物对不同代次CHO细胞的抑制率是否有显著差异。结果表明:随着细胞代次的升高,细胞的形态、胞浆折光性、边界、排列均发生了改变;同种受试物对CHO细胞的抑制率随细胞代次的升高呈上升趋势;中性红比色法较MTT比色法对细胞的衰老更灵敏。为了提高细胞毒性试验结果的可重复性以及可比性,细胞代次是需要控制的重要因素。
The effects of CHO cell continuous passage culture on the results of cytotoxicity test were studied in this paper. CHO Cell morphology was observed. Neutral red assay and MTT assay were used to examine whether the cell inhibition rates had a significant difference in different passages. The results showed that mor- phology,refractivity,boundary and arrangement of CH0 cell changed with the passage increasing. The higher cell passage numbers,the CHO cell inhibition rate was higher caused by the same inhibitor. Moreover,the neu- tral red assay was more sensitive to cell aging than MTT assay. To increase the repeatability and comparability of cytotoxicity test,cell passage was the important factor of quality control.
