摘要
目的观察中药生地黄免煎颗粒对神经生长抑制因子Nogo-A蛋白表达的影响。方法将实验大鼠分为用药组、模型组和假手术组,采用生地黄免煎颗粒溶液灌胃的方式作用于MCAO模型大鼠,分别在造模后第3、7、14 d 3个时间点采用Western Blot方法观察Nogo-A蛋白的表达。结果经MCAO造模造成脑缺血损伤之后,Nogo-A蛋白的表达出现升高趋势,且随时间呈现先上升后下降的趋势,在第7日表达最为活跃;MCAO造模后经生地黄免煎颗粒溶液灌胃干预以后,Nogo-A蛋白的表达水平在3个时间点上均较模型组降低,且呈现先升高后又缓慢恢复的趋势;相对于模型组,在造模后第14日,生地黄对Nogo-A蛋白的表达有明显的下调作用。结论生地黄免煎颗粒能够在一定程度上下调MCAO造模后引起的Nogo-A蛋白表达升高,有利于中枢神经缺血后的神经再生。
Objective: To observe the effects of Shengdihuang granules on the expression of neuron growth inhibitors Nogo-A protein in the rats following cerebral ischemia.Methods: All of the rats were randomly divided into 3 groups,i.e drug group(Group A),model group(Group B) and Sham-operated Group(Group C).The MCAO model rats were administered by gastrogavage with solutions of Shengdihuang granule.The expression of Nogo-A were recorded respectively in different time(3,7,14 day) following cerebral ischemia by Western Blot.Results: The MCAO model rats had the higher expression trend for Nogo-A protein following cerebral ischemia,Nogo-A ascended firstly,then decended with time.The highest expression of Nogo-A protein appeared in the seventh day.Compared with the model group,the expression of Nogo-A in the drug group rats decreased in different three time-points,had the trend of ascending first then recovering slowly.Compared with the model group,Shengdihuang had the significant down-regulation effects on the expression of Nogo-A in the rats following cerebral ischemia 14 days later.Conclusions: Shengdihuang granule can,to some extent,down-regulates the increasing expression of Nogo-A protein caused by MCAO model,and be helpful for nerve regeneration following cerebral ischemia in central nervous system.
