摘要
目的:探讨电针联合脂肪源性干细胞(ADSC)移植对大鼠缺血/再灌注损伤后海马区凋亡相关基因Livin和Caspase-3表达的影响。方法:采用线栓法复制大鼠大脑中动脉缺血再灌注模型,将动物随机分为模型组、电针组、ADSC移植组、电针+ADSC组。应用NSS法行神经功能损害评分,采用蛋白质印迹法检测海马区Livin和Caspase-3蛋白表达水平。结果:电针+ADSC组海马区PKH-26标记的细胞个数高于ADSC组(P<0.05)。电针+ADSC组NSS评分低于单独治疗组(P<0.05)。与模型组比较,电针+ADSC组和电针组Livin蛋白表达增高(P<0.05),Caspase-3蛋白表达下降(P<0.05)。结论:电针联合脂肪源性干细胞移植促进脑缺血大鼠神经功能恢复作用优于单独治疗组,其机制可能与电针通过上调海马区Livin表达,下调Caspase-3表达,抑制移植的ADSC凋亡有关。
Objective: To investigate the effect of electro-acupuncture (EA) combined with adipose-derived stem cell (ADSC) on Livin and Caspase-3in hippocampus after cerebral ischemia-reperfusion in rats. Methods: The rat model of middle cerebral artery occlusion was established in model, rats were randomly divided into model group, EA group, ADSC group and EA+ ADSC group. Neurological function score were observed by NSS, the expressions of Livin and Caspase-3in hippocampus were observed by using western blotting. Results : The number of labeled PKH-26in hippocampus of EA+ADSC group was higher than ADSC group. Compared with the single treated group, the NSS was significantly decreased in EA+ADSC group; Compared with the model group, the expression of Caspase-3in hippocampus was significantly decreased in EA group and EA+ ADSC group, however, the expression of Livin was increased. Conclusion: The combined treatment group has better effect than single treatment group on recovering neural function of cerebral ischemic rats, which may be related to up-regulating Livin expression, down-regulating Caspase-3expession, and then reducing apoptosis of ADSC.
