α-半水硫酸钙/辛伐他汀/万古霉素复合材料抗菌和细胞毒性作用的研究 被引量:3

Antibacterial properties and cytotoxicity of alpha half water calcium sulfate/simvastatin/vancomycin composite materials

摘要 [目的]研究不同浓度配比α-半水硫酸钙/辛伐他汀/万古霉素复合材料的体外抗金黄色葡萄球菌、大肠埃希菌和铜绿假单胞菌的作用,以及α-半水硫酸钙/辛伐他汀/万古霉素复合材料浸提液对新生小鼠成骨细胞的毒性作用。[方法]采用细菌生长抑制的定性和定量试验测定抑菌圈直径、抑菌率;MTT法细胞毒性试验测定α-半水硫酸钙/辛伐他汀/万古霉素复合材料浸提液体外对成骨细胞的相对增殖率及观察细胞形态学改变,并确定相应毒性等级。[结果]含5、10、20μg/mlα-半水硫酸钙/辛伐他汀/万古霉素复合材料对新生小鼠成骨细胞作用呈浓度-效应关系,对金黄色葡萄球菌和大肠埃希菌的抑菌圈平均直径分别为28、30、33mm和24、23、25mm,对铜绿假单胞菌没有抑菌作用。[结论]α-半水硫酸钙/辛伐他汀/万古霉素复合材料浸提液基本无细胞毒性,细胞相容性好,具有良好的抗菌效果,并能促进成骨细胞增殖和分化。 [ Objective ] Escherichia coli and staphylococcus aureus, the pseudomonas aeruginosa by alpha half water calcium sulfate/simvastatin/vancomycin composite materials, and the cytotoxicity of new osteoblast cells in mice by in vitro were studied. [ Methods ] With the qualitative and quantitative of bacteria inhibition growth experiments, the diameters and antibac- terial rates of Staphylococcus aureus ATCC25923, escherichia coli ATCC25922, pseudomonas aeruginosa ATCC27853 were determined under the affection of contained different concentrations of alpha half water calcium sulfate/simvastatin/vancomycin composite materials. The cytotoxicity tests of the alpha half water calcium sulfate/simvastatin/vancomycin composite materials on the new osteoblast cells in mice was made to evaluate the saftywith MTTcytotoxicity assay. [ Results] Containing 5, 10, 20 μg/ml alpha half water calcium sulfate/simvastatin/vancomycin composite materials for newborn mice osteogenesis cell concen- tration- effect relations on staphylococcus aureus and escherichia coli bacteriostatic circle diameter were 28, 30, 33 mm and 24, 23, 25 mm, including 0. 2%, 0. 4%, 8%, 1.0%. To pseudomonas aeruginosa ATCC27853 had no bacteriostasis. [ Conclusion] The alpha half water calcium sulfate/simvastatin/vancomycin composite material leaching solution of toxic reaction level in 2 below level, are the basic of cell toxicity, they have good antibacterial effect, cells with good compatibility, and can promote bone cell proliferation and differentiation.
