重组白介素-12对病毒性心肌炎小鼠IL-6、IL-10的影响 被引量:2

The change of IL-6 and IL-10 in mice with viral myocarditis treated by recombinant interleukin-12

摘要 目的检测重组白介素-12(rmIL-12)治疗小鼠病毒性心肌炎过程中血清白介素-6(IL-6)及(IL-10)水平,探讨rmIL-12的作用机制。方法 200只Balb/c小鼠随机分成6组:空白对照组、病毒对照组、药物对照组,以及rmIL-12小剂量治疗组、中剂量治疗组、大剂量治疗组。应用酶联免疫吸附法检测各组小鼠血清IL-6及IL-10水平,苏木精-伊红(HE)染色检测心肌病理改变。结果不同治疗组之间和治疗后不同时间,IL-6及IL-10值均有变化。在实验的第5及第10天,IL-6及IL-10的血清水平随着应用rmIL-12量的增加而增加。各治疗组IL-6血清水平在第5天达到高峰,在第15天各组IL-6血清水平均降低到接近空白对照组水平。IL-10在第10天达到高峰,在第15天各组间IL-10的血清水平有所下降,但尚未恢复到空白对照组水平。除空白组外,各组心肌均有炎症细胞浸润和变性坏死,病变最轻的为中剂量治疗组。结论适量应用rmIL-12治疗病毒性心肌炎,可通过调节Th1/Th2细胞因子平衡,使心肌病理改变减轻,而起到治疗作用。 Objective To detect the level of interleukin (IL) -6 and IL-10, and study the therapeutic mechanism of re combinant IL-12. Methods A total of 200 Balb/c mice were randomly divided into six groups, blank control group, the virus control group, drug control group, rmIL-12 small-dose treatment group, medium-dose treatment group and high-dose treat ment group. ELISA was used to detect the serum IL-6 and IL-I0 levels, and HE staining was used to observe myocardial pathological changes. Results There were significant differences of serum IL-6 and IL-10 levels among different treatment groups, and there were also significant differences of serum IL-6 and IL-10 levels at different time points after treatment. Five days and ten days after the treatment, the serum levels of IL-6 and IL-10 increased as therapeutic dosage of recombinant IL- l2 increased. Five days after the treatment, the serum level of IL-6 peaked. After 15 days of the treatment, the serum level of IL-6 decreased to the level of the blank control group. Ten days after the treatment, the serum level of IL-10 peaked. Fifteen days after the treatment, the serum level of IL-10 of each group decreased, but was still above the level of the blank control group. Myocardial pathological change was slighter in medium-dose treatment group than in the other treatment groups except the blank control group. Conclusions Appropriate amount of recombinant IL-12 can ameliorate the myocardial pathological changes and treat viral myocarditis through adjusting the balance of Thl and Th2 cytokines.
