摘要 目的探讨人轮状病毒(HRV)感染后腹泻的新生大鼠肠道上皮中Toll样受体2/4(TLR2/4)mRNA和蛋白水平的表达情况。方法 36只7日龄新生SD大鼠,随机分为实验组和对照组,每组18只。实验组于10日龄给予3×106PFU HRV灌胃,观察大鼠的腹泻程度和体质量变化,分别于感染后第1、5、9天处死,留取小肠标本,通过实时定量PCR和Western-blot分别检测肠道上皮细胞中TLR2和TLR4的mRNA和蛋白表达。结果 HRV感染后的新生大鼠出现不同程度的腹泻、体质量增长缓慢,与对照组比较差异有统计学意义(P<0.05);在第5天和第9天,实验组大鼠肠道上皮细胞TLR2和TLR4 mRNA和蛋白水平的表达量均较对照组增多,差异有统计学意义(P<0.05);实验组大鼠在第5天的表达量较第1天和第9天增多,差异有统计学意义(P值均<0.05)。结论 TLR2和TLR4 mRNA和蛋白水平在HRV感染后腹泻大鼠肠道上皮的表达量增多,提示先天性固有免疫与HRV感染后腹泻的发病机制存在潜在的相关性。 Objective To investigate the mRNA and protein levels of Toll like receptor 2 and 4 (TLR2/4) in intestinal epithelial cells of neonatal rats with rotavirus infection. Methods Thirty-six 7-day-old Sprague Dawley rats were randomly divided into control group and rotavirus infection group. Each group had eighteen rats. Rotavirus infection group were inocu lated with 3xlO6 PFU HRV on day 10. The extent of diarrhea and body weight were observed. On day 1, 5, 9 after infection, six rats were sacrificed. The intestine tissues were collected. The mRNA and protein'expression of TLR2/4 were detected by real-time PCR and western-blot. Results Compared with control group, various extent of diarrhea was observed and the body weight gain was slower in infected neonatal rats. The difference was statistically significant (P〈0.05). Compared with control group, the mRNA and protein expression of TLR2/4 in rotavirus infection group on day 5 and 9 were increased. In rotavirus infection group, the mRNA and protein expression of TLR2/4 on day 5 were higher than those on day 1 and 9. Conclusions The mRNA and protein expression of TLR2/4 were increased in intestinal epithelial cells of rotavirus infection rats. It indi- cated that the innate immune system was potentially involved in the pathogenesis of this disease.
