摘要 目的:探讨辣椒素诱导人结肠癌SW480细胞凋亡的分子机制。方法:不同浓度辣椒素处理SW480细胞,CCK-8细胞计数试剂盒法检测生长抑制作用,流式细胞术检测早期凋亡,RT-PCR和Western blot方法检测细胞HSP27、casepase-3、casepase-9 mRNA及蛋白水平表达。结果:辣椒素可显著抑制SW480细胞生长,作用呈时间、剂量依赖性,并可促进细胞早期凋亡,下调HSP27、上调casepase-3、casepase-9 mRNA和蛋白水平的表达。结论:辣椒素可通过下调SW480细胞HSP27表达、上调casepase-3及casepase-9表达,而起到诱导细胞凋亡、抑制细胞生长的肿瘤抑制作用。 Objective To investigate the apoptotic effect of capsaicin on human colon cancer cell line SW480. Methods SW480 cells were treated with capsaicin at various concentrations for different durations. Cell growth was determined by CCK-8 method.Flow cytometry was applied to measure the early apoptotic rate, so was RT-PCR and Western blot to detect the protein expressions of HSP27,caspase-3,and caspase-9 genes. Results Capsaicin suppressed the growth of SW480 cells markedly and induced apoptosis in a time and dose- dependent manner. RT-PCR and Western blot assay showed that capsaicin downregulated the expressions of HSP27 and upregulated the expressions of caspase-3 and caspase-9. Conclusion Capsaicin inhibits the pro- liferation of SW480 cells. Apoptotic induction may be the main antineoplastic mechanism, which is mediated by altering apoptosis-related genes through downregulation of Hsp27 and upregulation of caspase-3 and caspase-9 proteins.
