摘要
目的采用国产激流式生物反应器培养表达HBsAg的重组CHO细胞。方法分别应用美国NBS公司的Celligen310型生物反应器和国产AP20SC型激流式生物反应器(单个细胞培养袋和4个细胞培养袋)连续灌流培养重组CHO细胞,连续培养35 d,每24 h收获1次,采用ELISA法测定细胞收获液中HBsAg的滴度。结果 Celligen 310型生物反应器培养重组CHO细胞分泌的HBsAg在2.0μg/ml水平上可持续8 d,至第18天达最高水平(2.16μg/ml),第35天收液中HBsAg含量为1.12μg/ml;国产AP20SC激流式生物反应器单个细胞培养袋培养重组CHO细胞分泌的HBsAg在2.0μg/ml水平上可持续9 d,第18天达最高水平(2.20μg/ml),第35天收液中HBsAg含量为1.08μg/ml,与Celligen 310型生物反应器相比,收液中HBsAg含量在2.0μg/ml水平持续时间无明显差异;国产AP20SC激流式生物反应器4个细胞培养袋培养重组CHO细胞分泌的HBsAg在2.0μg/ml水平上可持续13 d,与该生物反应器单个细胞培养袋相比明显延长,第21天达最高水平(2.14μg/ml),第35天收液中HBsAg含量为1.5μg/ml,明显高于单个细胞培养袋培养的细胞。结论国产AP20SC型激流式生物反应器可代替进口生物反应器对传代细胞进行大规模培养,用于人用疫苗的生产。
Objective To culture recombinant CHO cells for expression of HBsAg in domestic riptide bioreactor.Methods Recombinant CHO cells were cultured by continuous perfused culture for 35 d in imported Celligen 310 bioreactor and domestic AP20SC riptide bioreactor with one and four culture bags respectively.The culture fluid was harvested every 24 h and determined for HBsAg titer by ELISA.Results The secretion level of HBsAg in recombinant CHO cells was 2.0 μg / ml within 8 d after culture in Celligen 310 bioreactor,and reached the maximum of 2.16 μg / ml on day 18,which was 1.12 μg / ml in the harvested culture fluid on day 35.However,the secretion level of HBsAg in recombinant CHO cells cultured in domestic AP20SC riptide bioreactor with a single culture bag was maintained at 2.0 μg / ml for 9 d,and reached the maximum of 2.20 μg / ml on day 18,which was 1.08 μg / ml in the harvested culture fluid on day 35.The maintenance time of HBsAg level at 2.0 μg /ml in the harvested culture fluid in domestic AP20SC riptide bioreactor showed no significant difference with that in Celligene 310 bioreactor.However,the secretion level of HBsAg in recombinant CHO cells cultured in domestic AP20SC riptide bioreactor with four culture bags was maintained at 2.0 μg / ml for 13 d,which was prolonged significantly as compared with that in bioreactor with a single culture bag,then reached the maximum of 2.14 μg / ml on day 21,and was 1.5 μg / ml in the harvested culture fluid on day 35,which was significantly higher than that in bioreactor with a single culture bag.Conclusion Domestic AP20SC riptide bioreactor may be applied to large-scale culture of subcultured cells for production of vaccines for human use instead of imported bioreactor.
