摘要
目的 研究枸橼酸乙胺嗪(DEC)对豚鼠离体气管条平滑肌收缩反应的影响及其机制。方法 利用离体方法,取乞管条放入37℃浴管的Krebs溶液中,分别在浴液中加入药物,测定KCl和5-羟色胺(5-HT)收缩气管条的作用和量效反应以及DEC和维拉帕米对KCI和5-HT作用的影响。结果 DEC能使KCI或5-HT引起的气管平滑肌收缩显著抑制,其IC50值分别为5.03mmol/L和9.54mmol/L。
Aim: To study the efffects and mechanisms of diethylcarbmazine citrate on contraction of isolated tracheal strips of guinea pigs. Methods: Using isolated tracheal strips technique. Each tracheal strip was placed in an organ bath filled with a Krabs solution of 37℃and various drugs was added respectively. It was determined that KCI or 5 -- HT induced contractile effect and its dose -- response curve on tracheal strips and DEC or Verapamil changed the effects of KCI or 5 -- HT. Results: The contractions induced by KCI or 5 -- HT in isolated tracheal of guinea pig were malkedly inhibited by diethylcarbmazine citrate(DEC), the IC50 value of DEC for contraction induced by KCI and 5 -- HT was 5 .03 mmol/L and 9. 54 mmol/L respectively. DEC caused ridhtward displacement of the dose -- response curse for KCI and 5 -- m, significantly depressed foe maxmal response, showing a nho -- competitive antigonism, the pD2 values were 1. 554and 1. 851 respectively, and DEC markedly inhibited the contractions induced by Ca2+ released form an intracellular store and extracellular Ca2+. Conclusion: The results suggest that DEC can significantly inhibit contraction of tracheal strips induced by KCI and 5 -- HT and its mechanism is rdsponsible calcium -- antagonistic effect of DEC.
