摘要
目的观察与ADAM-33相关的microRNA(miR)在卵清蛋白(ovalbumin,OVA)诱导的支气管哮喘小鼠模型中的表达。方法制备OVA诱导的支气管哮喘小鼠模型,检测支气管肺泡灌洗液(bronchoalveolar lavage fluid,BALF)中细胞数及建立组织病理验证模型。应用RT-PCR法检测支气管哮喘小鼠模型肺组织中ADAM-33 mRNA的表达差异。应用RT-PCR法检测支气管哮喘小鼠模型肺组织miR-140-5p、miR-148a、miR-138-1、miR-27b与对照组的表达差异。结果 OVA诱导的支气管哮喘小鼠模型组BALF中细胞总数及嗜酸性粒细胞数明显高于正常对照组。组织病理显示支气管哮喘模型组炎症细胞浸润明显高于正常对照组。ADAM-33 mRNA在OVA诱导的支气管哮喘小鼠模型肺组织中高表达。在支气管哮喘组,miR-140-5p的表达约为正常对照组的0.12倍;miR-148a的表达约为正常对照组的0.28倍;miR-138-1的表达约为正常对照组的0.35倍;miR-27b的表达约为正常对照组的0.22倍。结论 ADAM-33在OVA诱导的支气管哮喘小鼠模型中明显上调,相关的miR-140-5p、miR-148a、miR-138-1、miR-27b在OVA诱导的支气管哮喘小鼠模型中明显下调。
Purpose To investigate the expression of ADAM-33-related miRNA in a murine asthma model. Methods The number of cells in the bronchoalveolar lavage fluid (BALF) of OVA-induced murine asthma model was assessed. Expression of miRNA in the model was analyzed by realfime-PCR. Results The cell number was significantly higher in BALF from the murine asthma model than that from control. Enhanced eosinophilic infiltration in lung was observed in the murine asthma model. ADAM-33 mRNA was upregu- lated in the murinc asthma model. Compared to the control, miH-ld0-5p (about O. 12 fold) , miR-148a (about O. 28 fold), miR-138- l (about 0. 35 fold), and miR-27b (about 0. 22 fold) were downregulated. Conclusions ADAM-33 is significantly upregulated in the OVA-induced murine asthma model. Meanwhile, the down-regulation of ADAM-33-related miRNAs (miR-140-5p, miR-148a, miR-138-1 and miR-27b) is also observed in this model.
