PRK后兔角膜成纤维细胞分泌产生bFGF、TGF-β_1及其对成纤维细胞增殖的影响 被引量:7

摘要 目的 探讨在准分子激光屈光手术后角膜伤口愈合过程中 ,是否有生长因子b FGF及 TGF- β1 参与 ,两种生长因子对成纤维细胞增殖的作用及其相应抗体对这一过程的影响。方法 对 PRK后兔角膜成纤维细胞原代培养 ,观察有否内源性生长因子 b FGF及 TGF-β1 的作用 ,内源性及外源性生长因子 b FGF及 TGF-β1 对细胞增殖的影响及相应抗体对抗原的中和作用。结果 在 PRK后兔角膜伤口愈合过程中成纤维细胞分泌产生b FGF及 TGF- β1 ,并刺激细胞的增殖 ,相应抗体可阻断这一生物效应。结论  (1)准分子激光屈光手术后角膜愈合过程中成纤维细胞分泌产生生长因子 b FGF及 TGF- β1 ;(2 )两种生长因子对角膜成纤维细胞的增殖具有刺激作用 ;(3)相应抗体可阻断 b FGF及 TGF- β1 对细胞增殖的刺激作用。本研究结果提示抗 b FGF抗体及抗 TGF- β1 抗体作为准分子激光屈光手术后选择性用药的可能性 ,可防止或减少屈光回退和角膜上皮下混浊等两个主要并发症 。 Objective To study whether bFGF and TGF 1 were involved in the corneal wound healing after excimer laser refractive surgery,the role of the two growth factors on fibroblast proliferation,and the effect of anti bFGF antibody and anti TGF β 1 antibody during this process.Methods A primary culture of rabbit corneal fibroblast was established to observe if there were endogenous bFGF and TGF β 1,the effect of the two growth factors on cell growth,and the effect of anti bFGF antibody and anti TGF 1 antibody during this process.Results It approved that bFGF and TGF β 1 were produced by fibroblast,endogenous and exogenous growth factors bFGF and TGF β 1 stimulate the proliferation of rabbit corneal fibroblast after PRK,and the effect of bFGF and TGF β 1 were blockaded by anti bFGF antibody and anti TGF β 1 antibody.Conclusions (1)bFGF and TGF β 1 were produced by fibroblast during the corneal wound healing after excimer laser refractive surgery;(2)bFGF and TGF β 1 stimulate the proliferation of corneal fibroblast;(3)Anti bFGF antibody and anti TGF β 1 antibody can neutralize the effect of bFGF and TGF β 1.The result indicated that the two kinds of antibody can prevent and reduce the two main complications of regression and haze,and improve the effect of surgery.
