汉防己甲素对TGF-β1诱导L929细胞活化和分泌FN、α-SMA的影响

Effects of tetrandine on TGF-β1-induced activation and secretion of FN and α-SMA in L929 cells

摘要 目的研究汉防己甲素对转化生长因子β1(TGF-β1)诱导的L929细胞活化和分泌纤维连接蛋白(FN)、α-平滑肌肌动蛋白(α-SMA)的作用。方法体外培养L929细胞,分别以TGF-β1和汉防己甲素作用,观察细胞形态变化,测定FN及α-SMA mRNA、蛋白质表达水平。结果 TGF-β1诱导L929细胞72h,能显著上调FN及α-SMA mRNA、蛋白质表达;加入汉防己甲素后能显著抑制FN和α-SMA的表达。结论汉防己甲素能够抑制TGF-β1诱导L929细胞活化和分泌细胞外基质的作用。 Objective To invesgate the effects of tetrandine on TGF-β1-induced activation and secretion of fibronectin(FN) and a-SMA of L929 cells. Methods L929 cells(embroyo lung fibroblast) cultured in vitro were treated with TGF-β1 and tetrandine. The expressions of FN 及 a-SMA mRNA and protein were detected and the morphologic changes were observed. Results That TGF-β1 induced L929 cells for 72 hours upregulated the expressions of FN 及 a-SMA mRNA and protein,which were inhibited by adding tetrandine. Conclusion Tetrandine can inhibit TGF-β1-induced activation and secretion of L929 ceils.
