摘要
利用元基因组技术,直接提取印度洋深海浓缩海水的总DNA,利用细菌和古菌的通用引物扩增16SrRNA片段并建立文库,在对阳性克隆进行酶切验证的基础上,广泛大量地挑取单克隆进行测序,应用分析软件和数据库资料,研究印度洋深海微生物的多样性,结果发现了3个不同细菌的OTU.
This paper uses metagenome technology, and extracts the total DNA of the India Ocean's deep-sea water directly. The universal primers for bacteria and archaea is used to amplify the 16SrRNA fragment and establish the library. The monoclonal is selected widely for sequencing through the positive clones which are verified by enzyme digestion. The analysis software and database information are applied to study the diversity of the Indian Ocean deep-sea microorganisms, and the result is lhat three different bacteria's OTU are found.
