喉鳞癌组织KAI1和COX-2表达及相关性研究 被引量:1

Expressions and relationships between KAI1 and COX-2 of laryngeal squamous cell carcinoma

摘要 目的:了解KAI1和COX-2在喉鳞癌组织中表达的情况,探讨两者在喉鳞癌组织中表达的意义及相关性,以及在喉鳞状细胞癌发生、发展及浸润转移过程中的作用。方法:应用免疫组织化学(SP)法分别检测45例喉鳞状细胞癌的咽喉黏膜组织和10名正常体检者咽喉黏膜组织中KAI1和COX-2的表达水平,分析两者的表达水平及强度与相关临床指标之间的关系,并分析这两个基因之间在喉鳞状细胞癌中表达的相关性。结果:KAI1基因在喉鳞癌细胞中的表达低于在正常咽喉黏膜组织中的表达,χ2=4.11,P=0.043;在喉癌组织中表达的阳性率与年龄、性别、分化程度、临床分期及是否有淋巴结转移均无关(P值均>0.05),但其阳性表达强度与临床分期及是否有淋巴结转移有显著相关性,P值均<0.05。COX-2基因在喉鳞癌组织中高表达,在正常咽喉黏膜组织中低表达,差异有统计学意义,χ2=15.36,P=0.000,在喉鳞癌组织中表达的阳性率与性别、年龄及分化程度无关(P值均>0.05),而与临床分期及是否有淋巴结转移有显著相关性,P值均<0.05。KAI1和COX-2在喉鳞癌组织中的表达呈负相关,r=-0.30,P=0.045。结论:KAI1和COX-2在喉鳞癌的发生、发展过程中具有一定抑制和促进作用,可作为今后预测肿瘤细胞转移潜能的指标。 OBJECTIVE: To explore the expressions of the KAI1 and COX-2 in laryngeal squamous cell carcinoma, and the relevance of the two discussed with lymph node metastasis. METHODS: The expressions of KAI1 and COX-2 were detected by immunohistochemistry(SP) in 45 cases confirmed by pathology in patients with laryngeal squamous cell carcinoma of the throat mucosa and 10 normal volunteers. To analyze the expressions of the level and intensity of clinical features associated with the cases and the correlation of these two genes in laryngeal squamous cell carcinoma. RESULTS: KAI1 in laryngeal squamous cell carcinoma was lower than that in normal mucosal tissue of the throat (X2=4. 11, P=0. 043 ). There was no significant relationship in age,gender, degree of differentiation, clinical stage and lymph node metas tasis, but the intensity of expression with clinical stage and lymph node metastasis was significant correlated (P〈0.05). COX-2 gene in laryngeal squamous cell carcinoma was highly expressed while low expression in normal mueosa of the throat, the difference was significant (X2 : 15.36,P=0. 000). The expression of COX-2 in laryngeal squamous cell carci- noma was closely related to clinical stage and lymph node metastasis (P(0. 05) ,but not to gender,age and histological grade. KAI1 and COX-2 in laryngeal squamous cell carcinoma was contrarily correlated ( r= - 0.30, P = 0. 045 ). CON CLUSION : KAI1 and COX-2 have a certain inhibitory and promote effect in occurrence and the development process of la ryngeal squamous cell carcinoma,that may be used as detection of tumor cells and the metastatic potential of one of the in dicators.
