8号染色体四个单核苷酸多态性与中国前列腺癌风险的关联性研究 被引量:2

A study of chromosome 8q24 and 8q21 association with prostate cancer in Chinese population

摘要 探讨8q24SNP(rs6983561,C)、SNP(rs13252298,T)、SNP(rs12543663,T)和8q21SNP(rs1512268,T)四个风险等位基因位点与北京居民前列腺癌(prostarecancer,PCa)的关联,了解PCa患者基因型和表型关系。方法收集124例PCa患者遗传、膳食习惯和临床表型,对124名PCa患者和138名正常对照者的8q24SNP(rs6983561,C)、SNP(rs13252298,T)、SNP(rs12543663,T)和8q21SNP(rs1512268,T)四个等位基因位点进行比较,并进行病例组的基因型-表型分析。结果两组间相比,rs6983561(C)风险等位基因频率差异无统计学意义(病例组61.7%,对照组68.2%;P=0.12);rs6983561(T)等位基因及基因型在两组间分布差异无统计学意义(病例组87.1%,对照组92.0%;P=0.06);rs1512268(T)风险等位基因频率差异无统计学意义(病例组74.0%,对照组71.9%;P=0.60);rs12543663(T)风险等位基因频率差异无统计学意义(病例组71.7%,对照组72.4%;P=0.86)。数量性状分析,rs1512268的GG风险基因型与AG或AA基因型相比,携有GG的患者发病与患者饮酒的酒精度数有关(P=0.01)。结论 8q24区rs6983561(C)、rs13252298(T)、rs12543663(T)和8q21rs1512268(T)风险基因和中国北方人PCa易感性之间可能不存在相关性。 Objective to explore the correlation between chro-mosome 8q24:rs6983561(C)、rs13252298(T)、rs12543663(T)and8q21 rs1512268(T) and prostate cancer(PCa) in Beingjing residents,and to know the correlation between genotype and phenotype in PCa patients.Methods PCa patients phenotype wer-e collected inclding clinical,genetic,dietary habits,hobbies and blood samples.chromosome 8q24:rs6983561(C)、rs13252298(T)、rs12543663(T)and 8q21 rs1512268(T) compared the allele distribution between 124 PCa and 138 healthy control age matched patients.The genotype and phenotype analysis was c-onducted in the 2 groups.Resuts Between the case and control groups,the rs6983561,C of chromosome 8q (case 61.7%%,control 68.2%),the rs13252298,T of chromosome 8q (case 87.1 %,control 92.0%),rs1512268,T of chromosome 8q (case 74.0 %,control 71.9%),rs12543663,T of chromosome 8q (case 71.7 %,control 72.4%) all were not significantly associated with PCa in the study sample(P0.05).Quantitative trait analysis showed that the Drinking alcohol of rs1512268 risk genotype(G/G,37.67±20.00) degrees in PCa patients was significantly higher(P=0.012) than the Drinking alcohol of other genotypes carriers(A/G and A/A,32.20±17.413) degrees.Conclusion the rs6983561(C)、rs13252298(T)、rs12543663(T)and rs1512268(T) variation in chrmosome 8q are not possibly associated with PCa susceptibility,suggesting that the genes might not associa8q24 ted with PCa in a northern Chinese population but might associated with PCa in other population.
