摘要
目的探讨姜黄素对矽肺大鼠肺组织水通道蛋白-1(AQP-1)表达的影响及意义。方法雄性Wistar大鼠40只,随机分为正常对照组、生理氯化钠溶液组、矽肺模型组、姜黄素组和溶剂对照组。后3组均一次性气管内给予质量浓度为50 g/L的矽尘混悬液1 ml,生理氯化钠溶液组一次性气管内注入等体积的生理氯化钠溶液。染尘第2天,姜黄素组给予质量浓度为20 g/L姜黄素300 mg.kg-1.d-1(0.5%羧甲基纤维素钠溶解),溶剂对照组给予质量分数为0.5%羧甲基纤维素钠15 ml.kg-1.d-1。于第29天处死动物,采用免疫组织化学、免疫印迹法、实时荧光定量聚合酶链反应法检测大鼠肺组织AQP-1及mRNA表达。结果矽肺模型组、溶剂对照组肺组织中AQP-1表达较正常对照组和生理氯化钠溶液组减低(P<0.05),姜黄素组肺组织AQP-1表达较矽肺模型组增强(P<0.05)。结论姜黄素可使大鼠矽肺模型肺组织中AQP-1表达增强,提示姜黄素可能通过影响AQP-1的表达,在矽肺的发病过程中具有一定的保护作用。
Objective To explore the effect of curcumin on the expression and the significance of aquaporin-1 in lung tissue of silicotic rats. Methods 40 healthy male Wistar rats were randomly divided into 5 groups: a control, a saline control, a silico- tic model, a curcumin and a solvent control group. The groups of silicotic model, curcumin and solvent control were intratrachea- lly given silica (50 g/L) while saline control group was given the same dose of saline. 1 day later, curcumin group was given curcumin (300 mg/kg, dissolved in 0.5% sodium carboxymethyl cellulose by 20 g/L) per day and solvent control group was given 0.5% sodium carboxymethyl cellulose ( 15 ml/kg) per day. Immunohistochemistry, Western blot and Real-time PCR were used to detect distribution and expression of AQP-1 protein and mRNA in lung tissue of rats exposed to silica. Results The pro- tein and mRNA expression of AQP-1 in lungs of silicotic model rats was decreased, compared with those of the normal and the saline control ( P 〈 0. 05 ). The protein and mRNA expression of AQP-1 in lungs of curcnmin treated rats was increased, com- pared with those of silicotic model rats (P 〈 0. 05 ). Conclusion The expression of AQP-1 increased in curcumin group, which suggested that curcumin may play a protective role in the progress of silicosis by influencing the expression of AQP-1.
