细胞色素P4501B1在子宫内膜癌组织中的表达与ER相关

EXPRESSION OF CYTOCHROME P4501B1 IN HUMAN ENDOMETRIAL CANCER AND ITS ASSOCIATION WITH ER EXPRESSION

摘要 目的检测子宫内膜癌及正常子宫内膜中细胞色素P4501B1(cytochrome P450 1B1,CYP1B1)、儿茶酚氧位甲基转移酶(catechol-O-methyltransferase,COMT)的表达差异,探讨其在子宫内膜癌发生发展过程中的意义。方法采用免疫组织化学SP法对40例子宫内膜癌及35例正常子宫内膜组织切片进行染色,使用奥林巴斯公司DP70成像系统完成采图,应用Image-Pro Plus5.1图像分析系统对染色标本进行图像分析处理。结果子宫内膜癌组织腺上皮胞浆中CYP1B1的表达明显高于正常子宫内膜组织(P<0.05);而COMT的表达低于正常子宫内膜组织(P<0.05)。子宫内膜癌组织中CYP1B1表达随雌激素受体表达增强而增强(P<0.05)。CYP1B1与COMT表达在正常子宫内膜组织中存在正直线相关关系(r=0.384,P<0.05)。结论 CYP1B1在子宫内膜癌的发生过程中起重要的作用,其作用发挥依赖雌激素受体,而COMT起保护性作用。此2种酶共同维持机体内正常的雌激素代谢,防止过多致癌物的产生。 Objective To detect the expression of cytochrome P4501B1 (CYP1B1) and catechol- O- methyltransferase(COMT) in endometrial cancer and normal endometrium, and to explore their significance in occurrence and development of endometrial cancer. Methods Immunohistochemical method was used to detect the expression of CYP1B1 and COMT in 40 cases of endometrial cancer and 35 cases of normal endometrium. Olympus DP70 imaging system was used to collect image. Image - Pro PlusS. 1 image analysis system was applied on the stained specimens for image analysis: Results The expression of CYP1B1 was higher in endometrial cancer tissue than that in normal endometrium (P 〈 0.05), while the expression of COMT in endometrial cancer tissue was lower than that normal endometrium(P 〈 0.05). The expression of CYP1B1 in endometrial cancer became stronger with the increasing of estrogen receptor(ER) expression(P 〈 0.05 ). The expression of CYP1 B1 and COMT have positive linear correlation in normal endometrium( r = 0. 384, P 〈 0.05 ). Conclusion CYP1 B1 may play an important role in the occurrence of endometrial cancer depending on ER. COMT plays a protective role in the occurrence and development of endometrial cancer. The two enzymes maintain the normal estrogenmetabolism in the body, and prevent excessive carcinogens production.
