摘要
目的观察正天丸对偏头痛大鼠的预防作用及对脑组织中降钙素基因相关肽(CGRP)的影响。方法 SD大鼠随机分为正常组(生理盐水)、模型组(生理盐水)、西比灵治疗组(1mg/kg)、正天丸组(16.8mg/kg)。除正常组外,其余各组采用皮下注射硝酸甘油法复制大鼠偏头痛模型。观察其耳红情况、挠头时间等行为学指标后,采用酶联免疫吸附法(ELISA)测定各组大鼠脑组织中CGRP的含量。结果与正常组比,模型组大鼠出现耳红和连续挠头时间明显延后(P<0.05),CGRP的含量升高(P<0.05);经西比灵和正天丸预防给药后,偏头痛模型大鼠耳红和连续挠头的持续时间缩短,CGRP的含量降低,与模型组比较有统计学意义(P<0.05)。结论正天丸能明显改善大鼠偏头痛发作,可能与调节机体CGRP的释放有关。
Objective To study the effects of Zhengtian pills(ZTP) on calcitonin gene-related peptide(CGRP) and its protection against migraine.Methods Sprague-Dawle(SD) rats were divided into 4 groups:Normal group(saline),model group(saline),flunarizine group(1 mg/kg),and the ZTP group(16.8 mg/kg).Rat models with migraine were established by subcutaneously administration of nitroglycerin for 3 h,brain was collected,and CGRP was measured by enzyme linked immunosorbent assay.Results Compared with the normal group,the times of scratching and shaking head were increased and the contents of CGRP was decreased in model group(P0.05).After preventive administration of ZTP and the flunarizine,the duration of ear flaring got short,and the times of scratching and shaking head were decreased and the contents of CGRP in the rat brain were decreased.Conclusion ZTP could obviously decrease the attack of miglaine,which might be related with the regulation of CGRP in the rat brain.
