DETERMINATION OF MITOMYCIN C IN SERUM BY HPLC

本文采用HPLC法测定人血中丝裂霉素C。以异丙醇一氯仿(1/1)提取人血清,提取液以高纯度N_2在40℃水浴中吹干。以甲醇溶解残渣后进样测定。色谱条件:检测波长350nm,μ-Bondapak C-18柱(15cm×3.9mmID),流动相为磷酸盐缓冲液:甲醇(70:30v/v),流速1 ml/min,最低检测浓度0.05μg/ml,保留时间3.8-4.0min,挂温25℃。

A method is described for the determination of mitomycin C in serum using high performance liquid chromatography. The liquidliquid extraction with isopropanol -chloroform (1/1) is evaporated by pure nitrogen below 40℃to avoid decomposition of the drug. The residue is dissolved with pure methanol and then injeced into the chromatog-graphy system. Chromatography condition; UV detection at 350mm, u-Boudapak C 18(15 cm × 3.3mm ID) column, phosphate buffer; methanol=70:30 as morbile phase, folw rate I ml/min, the minimal detertable concentration is 0.05μg/ml, retention time is 3.8-4.0 min, temperature 25℃.
