苦豆子总碱对大鼠膀胱逼尿肌作用机制研究 被引量:2

Contraction Effects of Total Alkali Sophora Alopecuroids on Rat's Bladder Detrusor Muscle Strips in Vitro

摘要 目的:观察苦豆子总碱(total alkali sophora alopecuroids L,TASa)对大鼠膀胱离体肌条的作用及机制。方法:采用恒温灌流浴槽,记录膀胱逼尿肌收缩波的振幅、持续时间、收缩面积和频率,给予TASa(5、10、20、40、80 mg/L)观察其量效关系(r);将标本分为正常组(NS)、TASa、TASa+异搏定(10-7mg/L)、TASa+酚妥拉明(10-6mg/L)、TASa+苯海拉明(10-6mg/L)、TASa+消炎痛(10-6mg/L)和TASa+阿托品(10-6mg/L)七组探讨TASa兴奋逼尿肌的作用机制。结果:TASa能增强逼尿肌收缩的振幅、持续时间和收缩面积,并有量效依赖性(振幅r=0.95,t=5.43,P<0.05;持续时间:r=0.97,t=7.99,P<0.01;收缩面积:r=0.97,t=7.03,P<0.01),从TASa(20 mg/L)浓度开始可加快其收缩频率(P<0.01)。异搏定可抑制TASa兴奋逼尿肌的作用,而阿托品、苯海拉明、消炎痛和酚妥拉明则无影响。结论:TASa可能是激活Ca2+通道,升高胞浆内Ca2+触发逼尿肌的收缩。 Objective:To study the contraction effects of the total alkali of Sophora alopecuroids L(TASa)on rat's bladder detrusor muscle strips in vitro and the possible mechanisms.Method:The constant temperature perfusion of the bath machine was used to record amplitude,duration,contraction area and frequency of the contraction bladder detrusor muscle in vitro.TASa was divided into five doses groups(5,10,20,40,80mg/L),giving TASa with different doses to bladder detrusor muscle in vitro and the dose-effect relationship between muscle contraction and different doses of TASa was observed.Bladder detrusor muscle was divided into seven groups,including control group,TASa group,TASa+10-7mg/L verapamil group,TASa+10-6mg/L phentolamine group,TASa+10-6mg/L benzhydramine group,TASa+10-6mg/L indomethacin group,TASa+10-6mg/L atropine group.The contraction effects of TASa were investigated.Results:TASa can increase bladder detrusor muscle contraction amplitude,contraction duration,contraction area,and has an obvious concentration-dependent contraction(contraction amplitude r=0.95,t=5.43,P0.05;contraction duration r=0.97,t=7.99,P0.01;contraction area r=0.97,t=7.03,P0.01),and TASa(20mg/L)begains to accelerate the contraction frequency of bladder detrusor muscle.Verapamil can inhibit the bladder detrusor muscle contraction of TASa,however atropine,benzhydramine,indomethacin and phetolamine have no contraction effect.Conclusion:TASa may activate calcium channel,increase calcium flow to the interior of the cell and increase bladder detrusor muscle's contraction.
