目的探讨Claudin-1、Claudin-2、Claudin-3和Claudin-4在胃癌组织中的表达及意义。方法选取手术切除的胃癌患者72例，术后均经病理证实为胃癌，且未合并其它肿瘤疾病，应用免疫组化ABC法检测胃癌手术标本中Claudin-1、Claudin-2、Claudin-3和Claudin-4的表达。结果本样品中Clau—din-2的表达率为73．6％，Claudin4的表达率为44．4％，Claudin-1在腺癌中的表达水平明显降低，Clau—din-3和Claudin-4在淋巴转移阳性中的表达更低，Claudin-3在r13、T4中的表达明显降低，Claudin-3与其它Claudin蛋白质具有显著相关性，在患者生存分析里，Claudin-4与良好的预后有关。结论Claudin-3与Claudin-4可以作为胃癌的分子标志物，在淋巴侵袭的病例中检测Claudin-3与Claudin-4具有显著意义。

Objective To investigate the expression of Claudins in gastric carcinoma and analyze its significance. Methods Human gastric carcinoma tissue specimens were obtained from 72 gastric carcinoma patients between January 2008 and September 2009 at Department of General Suegery,first Affiliated Hospital, Gannan Medical university. All patients were diagnosed pathologically gastric cancer, and not associated with other tumor disease. The expression of Claudins were examined by immunohistochemistry. Results The expression rate of the Claudin-2 is 73.6% and claud^n-4 is 44.4%. The expression of claudin-1 was significantly lower in intestinal type adenocarcinoma. The expression of claudin-3 and claudin-4 were significantly lower in gastric carcinoma with lymphatic metastasis. The expression of claudin-3 was significantly lower in cases with an advanced T-stage （T3 and T4）. Claudin-3 has a significant correlation with the expression of the other Claudin protein, in survival analysis, the expression of Claudin-4 was related to good prognosis of gastric carcinoma. Conclusion Claudin-3 and Claudin-4 can be used as molecular markers to predict the biological behavior of gastric cancer. Detection of Claudin-3 and Claudin-4 in gastric carcinoma with lymphatic invasion has important significance.
