摘要
目的以哮喘小鼠模型B细胞产特异性IgE为指标,观察了T细胞及其亚群CD4+T细胞在哮喘发病机制中的调控作用。方法首先建立小鼠支气管哮喘模型,将小鼠的脾细胞制成悬液进行免疫细胞的分离。将T细胞及CD4+T细胞分别与B细胞共同培养,然后采用ELISA法对培养上清中特异性IgE的分泌水平进行检测。结果哮喘小鼠T细胞和CD4+T细胞分别与自身B细胞共同培养,上清中特异性IgE水平均明显高于B细胞单独培养;哮喘小鼠T细胞和CD4+T细胞分别与正常小鼠B细胞共同培养,上清中特异性IgE水平均明显高于B细胞单独培养;正常小鼠T细胞,分别与自身B细胞及哮喘小鼠B细胞共同培养,上清中特异性IgE水平与B细胞单独培养无差别。结论哮喘发病过程中,T细胞及CD4+T细胞对特异性IgE的产生有正性调控作用。
Objective To observe the control function of the T cell and CD4+ T cell to the specific IgE producing of B cell in the asthmatic murine model. Methods At first,established the model of asthmatic mouse,and then detach the immunologic cell from the mouse's spleen.Then cultured together T cells and CD4+ T cells with B cells respectively.At last,detected the concentration of specific IgE by the ELISA. Results When culturing together,the T cells and CD4+T cells of asthma mouse with self-B cells,the level of specific IgE were higher than that of culturing B cells singly(P0.01,P0.01).When culturing together the T cells and CD4+T cells of asthma mouse with B cells of normal mouse,the level of specific IgE were higher than that of culturing B cells singly(P0.05,P0.01).When culturing together the T cells of normal mouse with self-B cells and B cells of asthma mouse respectively,there were no significant difference from culturing B cells singly in the level of specific IgE(P0.01,P0.01). Conclusions T cells and the CD4+ T cells have positive control function to the production of the specific IgE in the process of asthma pathogenesis.
