摘要
目的 为临床探索一种更简便安全的下肢动脉缺血性疾病的基因治疗途径。方法 建立兔后肢缺血动物模型 ,将体外构建的重组质粒 pSV VEGF16 5注射于缺血肌群 ,30d后行动脉造影并切取标本 ,测定毛细血管密度和毛细血管 /肌肉数比值。结果 经 pSV VEGF16 5治疗后 ,缺血肢体毛细血管密度和毛细血管 /肌肉数比值显著增加 ,小腿部位比大腿部位增加明显 ;动脉造影显示部分缺血肢体侧支动脉明显增多。结论 肌肉内原位注射 pSV VEGF16 5是一种简单有效的治疗基因转染途径 ,能显著增加缺血肢体侧支血管形成。
Objective To investigate an easier and safer method of gene therapy in treatment of ischemic arterial disease of the lower extremity. Methods Recombinant plasmid pSV VEGF165 constructed in vitro were directly injected into the ischemic hindlimb muscles in rabbit models. 30 days after the gene transfer, selective angiography was undertaken, the capillary density and ratio of capillary to muscle fiber were determined histochemically in the resected specimens. Results The capillary density and capillary to muscle fiber ratio were significantly higher in pSV VEGF165 transfected group than those in control group, which were particularly increased in shank of the hindlimbs. In angiography, collateral arteries were significantly increased in some ischemic hindlimbs of pSV VEGF165 transfected group than those of control group. Conclusion Direct intramusclar injection of pSV VEGF165 was a simple and efficient gene therapy method, which can significantly stimulate collateral vessels development in ischemic limbs.(Shanghai Med J, 2000, 23∶210 212)
