溶媒对四氧嘧啶诱导建立糖尿病小鼠模型的影响 被引量:5

摘要 目的考察溶媒生理盐水和柠檬酸缓冲液对四氧嘧啶诱导糖尿病小鼠模型成模率的影响。方法分别比较溶媒生理盐水和pH值为4.4的柠檬酸缓冲液对四氧嘧啶不同剂量一次性腹腔注射及两次腹腔注射在成模率、死亡率及成模后空腹血糖值的影响。结果以柠檬酸缓冲液为溶媒,低剂量四氧嘧啶给药成模优于同剂量以生理盐水为溶媒四氧嘧啶,但当四氧嘧啶一次性给药剂量高至200 mg/kg,以柠檬酸缓冲液为溶媒成模率降低,死亡率升高。结论以pH 4.4柠檬酸缓冲液溶液为溶媒1%四氧嘧啶100、80 mg/kg两次腹腔注射成模率最高。 Objective To investigate the effects of physiological saline and citric acid buffer as solvent on establishing di abetic model in mice.Methods Diabetic model was replicated with intraperitoneal injection of Alloxan physiological saline and pH 4.4 citric acid buffer as solvent by different dose and different administration times.The influences of different solvents were observed and compared by evaluating the model successful rate,mortality and fasting blood glucose.Results The experiment results showed that the solution of Alloxan in citric acid buffer was more available than that in physiologi cal saline to induce diabetic mice when the dose of Alloxan was small.But when alloxan was given with dose to 200 mg/kg one-time,model success rate reduced and mortality increased.Conclusion A better diabetic mice model can be gained by twice intraperitoneal injection of Alloxan in pH 4.4 citric acid buffer of 100 and 80 mg/kg.
