摘要 目的 :研究Ku70基因导入人肺癌细胞 (LCA)后的表达及意义。方法 :构建重组的正义和反义Ku70基因表达质粒载体分别导入LCA细胞。结果 :用Northern印迹杂交法证明Ku70正义、反义基因均可在LCA的mRNA中得到表达 ,用West ern blotting法去证明细胞中只有导入正义Ku70基因的蛋白表达 ,而导入反义Ku70基因的人肺癌细胞 ,无正义Ku70蛋白表达 ,结论 :正义Ku70基因的表达可以调节DNA的修复 ,反义Ku70基因的表达可以增强肿瘤细胞对放射线照射的敏感性。 Objective: To study the expression and its specification of the Ku70 gene transferred into a human's lung carcinoma cell line (LCA). Methods: Constructing recombinant sense or antisense Ku70 expression vectors (Ku70/pCAGGS) and transferring into LCA. Results: Those Ku70 genes were expressed in the mRNA of the LCA cell line by Northern blotting . Only the sense Ku70 gene was expressed in the protein of the LCA transferred the sense Ku70 gene, but the sense Ku70 gene wasn't expressed in protein of the LCA transferred antisense Ku70 gene by Western blotting.Conclusion:The sense Ku70 gene would regulate to repair.The antisense Ku70 gene increased the LCA 's radiosensitivity DNA. They may offer a new molecular biology way to treat cancer.
