摘要
目的观察鲤鱼汤对大鼠心肌缺血再灌注损伤后肾脏损害的影响,并探讨其作用机制。方法 Wistar大鼠50只,随机分为对照组(10只)、模型组(20只)、治疗组(20只),鲤鱼汤治疗组给予鲤鱼汤灌胃,每天1次,连续15d,对照组及模型组则给予0.9%氯化钠溶液灌胃。利用冠状动脉结扎法建立大鼠的心肌缺血再灌注损伤模型,检测相关生化指标;光镜下观察心、肾的病理变化;ELISA方法检测血清中炎性因子白介素-2(IL-2)、γ干扰素(IFN-γ)的浓度。结果与对照组相比较,模型组血清中肌酸激酶(CK)、IL-2、IFN-γ明显升高(P<0.05),而内生肌酐清除率(Ccr)明显降低(P<0.01),血尿素氮(BUN)、血肌酐(SCr)未见明显变化;与模型组相比较,治疗组血清中CK、IL-2、IFN-γ明显降低(P<0.05),Ccr明显升高(P<0.01)。结论大鼠心肌缺血再灌注损伤对肾脏造成轻微损害,而鲤鱼汤对肾损害具有保护作用,其机制可能与抑制炎性因子IL-2、IFN-γ有关。
Objective To investigate the effect of cyprinus carpio decoction to kidney damage after the myocardium ischemia/reperfusion injury in rats and probe the mechanism of protection.Methods Fifty Wistar rats were randomly divided into nomal group(10),model group(20) and treated group(20).Rats in treated group animals were administered with cyprinus carpio decoction respectively once daily via gastric tube,while rats in nomal group and model group were administered with an equivalent volume of distilled water in the same way for 15 days.A rat model of myocardium ischemia/reperfusion injury was established.The serum was isolated to examine blood biochemical parameters.The pathological changes were observed under light microscopes.Results Compared with the control group,model group serum creatine kinase(CK),IL-2,IFN ppar-gamma increased significantly(P 〈0.05),and the endogenous creatinine clearance rate(Ccr) significantly reduced(P 〈0.01),blood urea nitrogen(BUN),serum creatinine(SCr) did not see obvious change;Compared with model group,the treatment group serum CK,IL-2,IFN ppar-gamma significantly reduced(P 〈0.05),Ccr increased significantly(P 〈0.01).Conclusion Cyprinus carpio decoction shows protective effects on rat kidney damage after the myocardium ischemia/reperfusion injury.The mechanism of protection might be related with upinhibition of IL-2,IFN-γ.
