东北鹤虱提取物LEGPS-Ⅱ对小鼠巨噬细胞氧化损伤的保护作用

摘要 目的:研究东北鹤虱提取物LEGPS-Ⅱ对小鼠巨噬细胞氧化损伤的保护作用。方法:采用昆明种小鼠,培养腹腔巨噬细胞(MФ),实验分为5组:正常对照组;H2O2氧化损伤组;3个浓度的LEGPS-Ⅱ(30、60、120μmol/L)保护组。用MTT法测定细胞存活率,比色法测定细胞培养液中一氧化氮(NO)含量、诱导型一氧化氮合酶(iNOS)活力、总超氧化物歧化酶(T-SOD)活力、丙二醛(MDA)含量。结果:与正常对照组相比,经H2O2处理过的氧化损伤组,细胞存活率明显降低(P<0.01),NO的含量、iNOS的活力明显升高(P<0.01),T-SOD活力明显下降(P<0.01),MDA含量明显升高(P<0.01);与H2O2损伤组相比,给LEGPS-Ⅱ各组均显著提高细胞存活率(P<0.05,P<0.01);使细胞培养液中NO的含量、iNOS的活力显著降低(P<0.05,P<0.01),T-SOD活力显著提高(P<0.05,P<0.01),MDA含量显著降低(P<0.01),且呈现剂量依赖性。 Objective: To research the protective effects of Lappula Echinata Gilib extract (LEGPS-Ⅱ ) on murine peritoneal macrophage with oxidative damage. Methods- The peritoneal macrophages from KM mice were cultivated in this experiment. The macrophage were divided into 5 groups randomly: Control group; H202 group; 30, 60, 120 μmol/L LEGPS- Ⅱ groups. The viabilities of macrophage were detected using MTT method. Activity of total superoxide dismutase(T-SOD), inducible nitric oxide synthase( iNOS )and content of malondialdehyde (MDA) and nitric oxide(NO) in the culture medium were detected using colorimetric method. Results: Com- pared with control group, in the group treated with H202, the cell viabilities was decreased significantly (P〈0.01), the content of NO and the activity of iNOS were increased significantly (P〈0.01), the activity ofT-SOD was decreased (P〈0.01), the content of MDA were increased significantly (P〈0.01); Compared with H2O2 group, in LEGPS- Ⅱ groups, the cell viability was increased significantly (P〈0.05, P〈0.01), the content of NO and the activity of iNOS were decreased significantly (P〈0.05, P〈0.01), the activity of T-SOD was increased (P〈0.05, P〈0.01), and the content of MDA were decreased significantly (P〈0.01), with the concentration dependent. Conclusion: LEG- PS- Ⅱcan protect peritoneal macrophage with H2O2 damage in different stages through its antioxidant effects.
