摘要
目的观察脐带间充质干细胞(umbilical cord mesenchymal stem cells,UC-MSC)移植对胶原诱导型关节炎(CIA)大鼠血清白介素-1(IL-1)、IL-10、IL-17、IL-18表达水平的影响,以探讨UC-MSC移植对CIA大鼠炎症因子的调控作用。方法 90只SD大鼠随机分为空白对照组、模型组、UC-MSC治疗组,每组各30只;采用鸡Ⅱ型胶原加完全弗氏佐剂建立CIA大鼠模型;治疗前后采用关节炎指数(AI)评分对大鼠关节炎症进行评估;以酶联免疫吸附试验(ELISA)检测各组大鼠治疗前后血清IL-1、IL-10、IL-17、IL-18水平变化。结果模型组血清IL-1、IL-17、IL-18水平均明显高于同期空白对照组,而IL-10水平则低于同期空白对照组,差异均有统计学意义(均P<0.05);UC-MSC治疗组治疗后第1、3周时IL-1、IL-17、IL-18水平低于同期模型组,IL-10水平高于同期模型组,差异均有统计学意义(均P<0.05);至第5周上述指标与空白对照组比较,差异均无统计学意义(均P>0.05)。结论 CIA大鼠存在免疫功能紊乱,其促炎症因子水平显著增高,抑炎症因子降低;UC-MSC能下调CIA大鼠促炎症因子水平,而上调炎症抑制因子,减轻机体免疫性炎症反应,促进疾病缓解。
Objective To observe the immunomodulation effects of umbilical cord mesenchymal stem cells (UC-MSC) on the IL-1, IL-10, II,-17, IL-18 of rats with collagen induced arthritis (CIA). Methods 90 cases of SD rats were randomly divided into control group, model group and UC-MSC treatment group with 30 cases in each group; chicken collagen type 11 with complete Freund's adjuvant (CFA) were used to immunize and establish the CIA rat model; the arthritis index (AI) was used to estimate the rats' joint inflammation before and after the treatment; ELISA was used to detect the changes of IL-1, IL-10, IL-17 and II,-18 in the rates before and after the treatment. Results IL-1, IL-17 and IL-18 in model group were higher than those in control group, IL-10 in model group were lower than those in control group, the differences were all statistically significant (all P 〈 0.05); 1L-1, 11,-17 and IL-18 in UC-MSC treatment group 1, 3 weeks after the treatment were lower than those in model group in the same term, 1l,-10 in UC-MSC treatment group were higher those in model group in the same term, the differences were all statistically significant (all P 〈 0.05); the differences of the indexes abovementioned belween the UC-MSC treatment group and the control group were all not statislically significant (all P 〉 0.05). Conclusion CIA rats have immune disorders, the proinflammatory factor is higher than normal level and the inflammation in- hibitory laetor is lower than normal level and promote disease in remission.
