大孔吸附树脂分离富集九节茶浸膏中迷迭香酸的研究 被引量:3

摘要 目的考察大孔吸附树脂对九节茶浸膏中迷迭香酸的吸附性能和纯化效果,寻找分离富集迷迭香酸较优的技术工艺。方法以迷迭香酸的吸附量和解析率为指标,从6种不同类型大孔树脂中筛选出较好的HPD-400树脂;以动态实验中迷迭香酸转移率及所得干膏中迷迭香酸的含量对树脂分离富集的最佳工艺进行筛选。结果 HPD-400树脂分离效果最佳,采用HPD-400树脂从九节茶浸膏中分离富集迷迭香酸的较优工艺参数为:上样体积4BV,速度2BV/h,收集洗脱液3BV,洗脱溶剂为70%乙醇。按照此工艺,迷迭香酸的平均过柱转移率为88.70%,洗脱液浓缩干燥后,所得干膏中迷迭香酸平均含量为4.57%,较原九节茶浸膏提高了5.2倍。树脂经5次重复使用,分离效果无明显下降,可以重复使用。结论 HPD-400树脂可以用于分离富集九节茶浸膏中迷迭香酸,达到了"去粗存精"的目的,具有潜在的工业应用价值。 Objective To evaluate the absorption capability and enrichment effect of rosmarinic acid from Sarcandra Glabra Exteact on macroporous resin, and find a better method to separate and enrich. Methods HPD-400 was separated from six kinds of resins due to macroporous's absorption capacity and transfer ratio. Methods for resins'separation and enrichment choosen was based on rosmarinic acid recovery rate and the content of rosmarinic acid in the extract. Macroporous resin HPD-400 was more suitable for separation and enrichment of rosmarinic acid among six konds of resins selected with ab- sorption capacity and transfer ratio. The better methods was screened for rosmarinic acid recovery rate and the content of rosmarlnic acid in the extract. Results Macroporous resin HPD-400 was more effective. The optimum technology parame- ters were as follows: volume was 4BV, sample flow rate was 2BV/h, collected eluent was 3BV, and desorbed by 40% alco- hol. The average transfer ratio of rosmarinic acid was 88.70%, and the average content of rosmarinic acid in the extract was 4.57%. It was 5.2 times of the sarcandra glabra extract. After five times of reuse, the separation effect of the macroporous redin was not decreated obviously, and the macroporous Glabra Extract can be effectively separated and purified cation value in industry. redin could be recycled.
