摘要 目的探讨细胞间黏附分子非整合素蛋白3(CLEC4L)介导丙型肝炎病毒(HCV)对人胎盘滋养层细胞感染情况,观察相关抗体体外干预CLEC4L与HCV结合作用的初步研究。方法培养滋养层细胞设CLEC4L单抗组、甘露聚糖组,同时设对照组。HCV与CLEC4L作用之前,先分别加入CLEC4L单抗及甘露聚糖进行干预,同时以高滴度HCV RNA阳性血清与各组细胞共培养。经RT-PCR法检测HCV感染后细胞内HCV RNA含量的变化。结果 CLEL4L单抗组细胞内HCV RNA水平较低,甘露聚糖组显示类似结果。结论 CLEC4L能够介导HCV进入人胎盘滋养层细胞,CLEC4L单抗和甘露聚糖对HCV与人滋养层细胞的结合有一定阻断作用。 Objective To investigate the functional contribution of human C - type lectin domain family 4 member L(CLEC4L) to hepatitis C virus(HCV) infection of human placental trophoblast cells,and to explore the protective effects of CLEC4L antibodies in vitro.Methods Primary human trophoblast cultures were established and divided into three groups.Two groups were pre-treated(blocked) with CLEC4L monoclonal antibody and mannan respectively,and the third group was used as untreated control.All three groups were subsequently co -cultured with high - titer HCV RNA - positive patient serum.The amount of HCV RNA copies in infected cells of the three groups was measured by RT - PCR.Results Compared with untreated control cells,HCV RNA was significantly lower in cells pre - treated with the CLEC4L monoclonal antibody and the cells pre - treated with mannan.Conclusion Antibody blockade of CLEC4L suppresses HCV entry into trophoblasts. CLEC4L monoclonal antibody and mannan may represent potential therapeutic inhibitors of HCV infection.
