转基因MSC体内外荧光表达的对比研究

摘要 目的:探讨转基因MSC在体内外表达荧光是否存在差异及可能的影响机制。方法:用转基因MSC输注到受射线照射的雌性BALB/C小鼠体内,输注后1、7、14、21、28、35天处死小鼠留取胸腺左叶,制作快速冰冻切片,在荧光显微镜下观察荧光表达,同时对比观察体外培养转基因MSC的荧光表达情况。结果:小鼠体内的转基因MSC在输注后1天开始出现荧光,第7天达高峰,第35天消失;体外培养的MSC制备后10h可见绿色荧光表达,第7天后表达率最高,第28天基本消失。结论:转基因MSC体内荧光表达优于体外,其机制与微环境及培养方式有关。 Objective To study the difference about fluorescent expression of transgenosis MSC in vivo and in vitro and the possible mechanism. Methods Female BALB/C mice were irradiated with 60Co y-rays and injected MSC 2h later. Fluorescent expression of the mice's thymus frozen sections were observed under fluorescence microscope on the 1st,7th,14th,21st,28th,35th day after irradiation. At the same time, fluorescence expression of transgenosis MSC were observed in vitro. Results Fluorescence pathology revealed the fluorescent expression reached the peak on the 7th day, dissapeard on the 35th day in vivo, and reached the peak on the 7th day, disspeared on the 28th day in vitro. Conclusion The fluorescent expression of transgenosis MSC in vivo is better than expression in vitro, the mechanism maybe is related with the microenvironment and the way of culture.
